硫化氢供体抑制低氧大鼠尾加压素II表达

目的:探讨硫化氢(H2S)在低氧性肺血管结构重建形成中对肺动脉人尾加压素II(hU-II)表达的调节作用,方法: 24只雄性大鼠随机分为对照组(n=8)、低氧组(n=8)和低氧+NaHS组(n=8)。弹力纤维染色后计算肺动脉的相对中膜厚度(RMT),免疫组化法检测大鼠肺动脉中hU-II蛋白表达。结果:与对照组相比,低氧组大鼠肺小型肌性动脉RMT增加41%,肺动脉内皮细胞和平滑肌细胞hU-II蛋白表达分别增加49%、78%。与低氧组相比,低氧+NaHS组大鼠肺小型肌性动脉RMT减少36%,肺动脉内皮细胞和平滑肌细胞hU-II蛋白表达分别降低30.2%、29.3%。结论: H2S显著抑制低氧性肺血管结构重建大鼠肺动脉尾加压素II表达,该效应可能参与H2S对低氧性肺血管结构重建的调节作用。
Objective: In this experiment the effect of H2S on the expression of human urotensin-II (hU-II) in the pulmonary artery during the development of hypoxic pulmonary vascular structural remodeling was explored. Methods: Twenty-four male rats were randomly divided into control group (n=8), hypoxia group (n=8) and hypoxia+NaHS group (n=8). The relative media thickness (RMT) of the pulmonary small muscular arteries was measured by elastin staining technique. The expression of hU-II in pulmonary muscular arteries were detected by immunohistochemistry and calculated by semi-quantitative analysis method. Results: Compared with rats in the control group, RMT of pulmonary small muscular arteries increased by 41% for rats in hypoxia group (p < 0.01). Semi-quantitative analysis showed that the protein expression of hU-II in pulmonary arteries of endothelial cells and the smooth muscle cells was all increased by 49% and 78%, respectively for rats in hypoxia group ( p all <0.01). Compared with rats in the hypoxia group, RMT of the pulmonary small muscular arteries for rats in hypoxia+NaHS group decreased by 36%. The expression of hU-II in pulmonary arteries of endothelial cells and the smooth muscle cells was decreased by 30.2% and 29.3%, respectively for rats in hypoxia+NaHS group. Conclusion: H2S may exert the inhibitory effect on the hU-II expression in the pulmonary artery under hypoxia.
