Geminin is a small and multifunctional nucleoprotein, which play a role in cell cycle regulation, embryomic development and tumorigenesis. Here we identified two Geminin homologous genes in silkworm: BmGem1 and BmGem2. BmGem1 contains the Geminin's coiled-coil domain and localize to the nucleus. Moreover, BiFC and Co-IP confirmed that BmGem1 can forms a complex with BmCdt1 in the nucleus. To investigate the function of BmGem1, we silenced BmGem1 by RNA interference and overexpressed BmGem1 in silkworm cell. Depletion of BmGem1 led to rereplication and the formation of giant nuclei in cells. In addition, overexpression of BmGem1 affects cell cycle progression: cells accumulate in S phase and are unable to efficiently progress to mitosis. These data show that BmGem1 may bind to BmCdt1 to regulate the cell cycle of silkworm.?????
