PTENp1, as the ceRNA of PTEN, competitively regulates the target relationship between miR-21 and PTEN in oralsquamous cell carcinomas
目的：探讨PTENp1作为ceRNA对miR-21与PTEN靶向作用中的调控效应。方法：构建miR-21过表达载体及PTEN野生型（PTEN-MT）和突变型（PTEN-WT）真核表达载体，筛选si-PTENp1，采用双荧光素酶报告基因、qRT-PCR和western blot实验检测miR-21对PTEN的直接调控，以及PTENp1介导的miR-21对PTEN的间接调控。结果：采用测序及NCBI Blast分析提示miR-21过表达载体及及PTEN野生型（PTEN-MT）和突变型（PTEN-WT）真核表达载体构建成功。miR-21能够显著抑制PTEN-WT的荧光活性（P = 0.014）；PTEN-WT、miR-21和PTENp1共转染时，荧光活性显著升高（P = 0.0086），然而PTEN-WT、miR-21和si-PTENp1共转染时，荧光活性显著抑制（P = 0.0095）；PTENp1过表达能够在mRNA和蛋白水平显著缓解miR-21对PTEN的靶向抑制效应（P＜0.05）。结论：PTENp1能够吸附miR-21，抑制miR-21对PTEN的靶向调控作用。
Objective: To investigate the PTENp1 to regulate the target relationship between miR-21 and PTEN in oral squamous cell carcinoma. Methods: miR-21 overpression, PTEN-MT and PTEN-WT plamids were constructed and si-PTENp1s were screened. Furthermore, luciferase reporter assay, qRT-PCR and western blot were performed to evaluate the target relationship between miR-21 and PTEN with or without PTENp1. Results: miR-21 overpression, PTEN-MT and PTEN-WT plamids were successfully identified using sequencing analysis and NCBI Blast. miR-21 overexpression could significantly inhibit the luciferase activity of PTEN-WT（P = 0.014）. Furthermore the luciferase activities were significantly increased in HEK 293 cells cotransfected with PTEN-WT, miR-21 and PTENp1（P = 0.0086）, while the luciferase activities were significantly decreased in HEK 293 cells cotransfected with PTEN-WT, miR-21 and si-PTENp1（P = 0.0095）. PTENp1 overexpression attenuated the suppressive effects of miR-21（P＜0.05）. Conclusion: PTENp1 could sponge miR-21, and attenuate suppressive effects between miR-21 and PTEN.
