软X射线纳米CT综述

软X射线纳米CT是进行细胞成像,检测亚细胞结构和细胞器的理想工具。由于工作在"水窗"波段,蛋白质等生物成分对射线的吸收系数比水的吸收系数高一个量级,这种特性使得含水生物样品无需染色和脱水,只需冷冻处理就能实现高衬度成像。对细胞等样品进行快速冷冻形成玻璃态,能够保持样品鲜活时的结构状态,如此成像可以获得原生态的三维结构信息。近年来,软X射线纳米CT在细胞、病毒、药物研究等领域都得到了广泛的应用,其相关设备也越来越受重视和得到了长足发展。该文将对其做一个综述,介绍其基本信息和应用。
Soft X-ray nano-CT is a powerful tool to detect subcellular organelles. Photons within "water window" region are absorbed an order of magnitude more strongly by proteins etc. carbon-rich organic material than by water. This special nature also allowed cells to be imaged without chemical dehydration or staining. The only step required to prepare samples is plunging the sample into cryogenic liquid nitrogen for vitrification, which could maintain the structural as the alive cell. In recent years, the soft X-ray nano CT have been widely used for cells, viruses, pharmaceutical and other fields imaging. Its associated equipment attracts more and more attention and considerable progress has been made. This paper will give an overview of soft X-ray tomography and its applications.
