目的:研究乳酸铝对原代脉络丛上皮细胞LRP-1及其配体表达的影响,初步探索铝引起Aβ在脑内蓄积是否与LRP-1及其配体有关。方法:培养原代脉络丛上皮细胞,7天后用不同浓度(空白组、低剂量、中剂量、高剂量)乳酸铝染毒。用Western Blot方法检测LRP-1的表达,用RT-PCR方法检测LRP-1及其配体(α2m、Apoj、TTR)mRNA的表达。结果:中、高剂量组LRP-1蛋白的表达量较空白组的表达较低,差异有统计学意义(P<0.05)。LRP-1、α2m、Apoj、TTR mRNA相对表达量均低于空白组表达量,差异有统计学意义(P<0.05)。结论:铝引起Aβ在脑内蓄积机制可能与LRP-1及配体表达的下降有关。

Objective: In this study, we aim to investigate the effect of aluminum lactate on the expression of LRP-1 and its ligands in primary cultured choroid plexus epithelial cells (CPEC). Methods: We cultured CPECs from choroid plexus of SD rats aged 14 days. Different concentrations of aluminum lactate as control, low-, mid- and high-doses were added into the CPECs 7 days. The protein’s expression of LRP-1 were analysed by Western Blot. The genes’ expression of LRP-1, α2m, Apoj, TTR were quantified by real time PCR, Results: The proteins’expression of LRP-1 in the mid-dose, high-dose was significantly lower (P<0.05) than that in the control. The genes’expression of LRP-1, α2m, TTR, Apoj were significantly lower (P<0.05) than those in the control. Conclusion: The mechanism that aluminum lactate could induce accumulation of Aβin the brain maybe relate to the reduction in the expression of LRP-1, and its ligands of α2m, Apoj, TTR.
