STI571抑制PDGF诱导的A375细胞迁移的作用机制研究
STI571 inhibits PDGF-induced A375 cell migration
肿瘤细胞自身的迁移是肿瘤转移过程中极为重要的一个环节。因此,发现能够有效抑制肿瘤细胞迁移的药物并对它们的作用机理进行深入探讨是目前肿瘤研究的一个热点。在本研究中,我们检测了c-Abl激酶抑制剂STI571对PDGF(血小板生长因子)诱导的恶性黑色素瘤A375细胞迁移的抑制作用,并初步探讨了相关的作用机理。结果表明,PDGF能够有效诱导αvβ3整联蛋白介导的A375细胞迁移,PDGF的这种作用能够被STI571显著阻抑。STI571处理导致PDGF诱导的αvβ3整联蛋白和c-Abl激酶共极化的消失,这可能是因为c-Abl激酶抑制剂STI571能有效抑制c-Abl激酶和avβ3整联蛋白相互作用。因此,我们的研究为STI571在抗黑色素瘤方面的应用提供了初步的理论基础。
ancer cell migration plays an important role in metastasis. Thus, at present, researchers have paid considerable attention to exploit novel anti-migration agents and reveal the possible mechanisms of their anti-migration. In this research, we detect the effect of STI571, a c-Abl kinase inhibitor, on PDGF-induced human malignant melanoma A375 cell migration. The results indicated that STI571 dramatically prevented PDGF-induced, αvβ3 integrin-dependent A375 cell migration. Furthermore, STI571 treatment resulted in the disassociation of αvβ3 integrin and c-Ab kinase, which was responsible for the inhibition of their co-polarity. Taken together, these data underscore the potential of STI571 as an effective melanoma-targeting drug.
