活化Notch1信号对人舌鳞癌细胞生长的影响及作用机制的研究
Effecting of Active Notch1 on Cell Proliferation in Human Tongue Carcinoma Cells
目的 构建重组逆转录病毒表达载体pCLNRX-ICN,研究过度活化Notch1信号对人舌鳞癌细胞系Tca8113生长的影响,并对其作用机制进行探讨。方法 将人Notch1基因胞内区(intracellular domain of Notch,ICN)克隆至逆转录病毒表达载体pCLNRX中,采用lipofectamineTM 2000将pCLNRX-ICN与包装载体pCL-10A1共转染293T细胞,制备重组病毒上清,检测病毒滴度。通过逆转录病毒感染,将ICN基因导入并整合到人舌癌细胞Tca8113细胞的基因组中,使Notch1信号在细胞中过度活化。MTT法检测稳定表达Notch1(ICN)对Tca8113人舌癌细胞生长的影响, Western blot检测Notch1和细胞周期蛋白激酶(cyclin dependent kinase ,CDK) 抑制因子p21WAF1/CIP1的变化。 结果 成功构建重组逆转录病毒载体pCLNRX-ICN,通过共转染可获得具有较高滴度的重组逆转录病毒。过度活化Notch1信号可以抑制人舌癌细胞Tca8113的生长,并上调CDK抑制因子p21WAF1/CIP1蛋白的表达水平。 结论 过度活化Notch1信号可部分通过上调CDK抑制因子p21WAF1/CIP1蛋白的表达水平而抑制人舌癌细胞生长。
Objective To construct the recombinant retroviruses expressing the constitutively active form of Notch1(intracellular domain of Notch,ICN) and to explore their effects on the growth of human tongue carcinoma cells Tca8113. Methods Notch1 (ICN) genes were inserted into retrovirus expression vector pCLNRX. The expression vector pCLNRX-ICN and packaging vector pCL-10A1 were co-transfected into 293T package cells by lipofectamineTM 2000. The recombinant retroviruses were used to infect human tongue carcinoma cells Tca8113. After infected, proliferation of Tca8113 cells was observed by MTT assay.Notch1 and p21WAF1/CIP1expression was detected by Western blot. Results Recombinant retrovirus vector pCLNRX-ICN was successfully constructed. After infection of recombinant retrovirus, the growth of Tca8113 cells was inhibited significantly. Western blot showed that Notch1 signaling up-regulated p21WAF1/CIP1 expression. Conclusion Constitutively active at exaggerated levels could inhibit the growth of tongue carcinoma cells, at least partially, through induction of p21WAF1/CIP1 expression.
