家蚕未知基因Bm-P312的克隆表达及定位研究

从本实验室构建的家蚕蛹cDNA文库中筛选获得一条阅读框为870bp,编码289个氨基酸的基因,GenBank登陆号为:DN237560,在NCBI上对该序列进行同源性比对后初步确定其为家蚕中的一未知基因,并命名为Bm-P312(Bombyx mori-P312)。我们根据开放阅读框设计上下游引物,PCR扩增目的基因,构建pET-28a-Bm-p312重组质粒,转入大肠杆菌BL21(DE3)中IPTG诱导表达该融合蛋白并进行纯化,免疫新西兰兔制备多克隆抗体。以家蚕Bm5细胞进行免疫细胞化学实验,结果显示,Bm-P312蛋白均主要分布在细胞质中。利用荧光定量PCR的方法,对家蚕卵、五龄幼虫、蛹、蛾四个发育时期,以及五龄幼虫各组织中Bm-P312的mRNA转录水平进行比较,分析发现在蛹期和生殖腺、头部中转录水平较高。提取家蚕卵、五龄幼虫、蛹、蛾四个发育时期及五龄幼虫各组织的总蛋白,进行免疫印迹实验,结果显示,Bm-P312蛋白在家蚕的蛹期以及生殖腺、头部、丝腺中均有明显表达,转录和表达情况相对比较一致。我们推测Bm-p312可能在家蚕的生殖发育过程中发挥一定作用,对Bm-P312蛋白功能的了解还待深入研究。

We got a novel gene with an open reading frame(ORF)of 870bp and encoding 289 amino acids from the silkworm pupae cDNA library constructed by our laboratory. The GenBank accession number is DQ237560.,After blasting the sequence homology in the NCBI datadase, we identified it was an unknow gene in the Bombyx mori andnominated the gene as Bm-P312(Bombyx mori-P312).According to the ORF of Bm-P312, two primers were designed to obtain the coding region of Bm-P312 gene. We ampifyed the Bm-P312 gene and constructed the rocombinant plasmid pET-28a-Bm-p312, The recombinant plasmid was transformed into E.coli BL21(DE3). and the recombinant protein was expressed in the E.coli BL21(DE3) induced by IPTG, then the recombinant protein was purified and immuned the rabbit to get antiserum.we located the Bm-P312 by Immunocytochemistry in Bm5 cells , the results indicated that the protein mainly existed in the cytoplasm.We analyzed the transcription level of Bm-P312 mRNA in different stages of silkworm and different tissues of the fifth instar larva. These results suggested that Bm-P312 had a high transcription level in silkworm’s pupa and seminal glands and brain . The total proteins was extracted from different stages of silkworm and some tissues of the fifth instar larvae, such as brain, epidermis, silk gland, midgut, Malpighian tubule, fat body, stigma and analyzed by Western blotting. The results showed that Bm-P312 highly expressed in pupa、seminal glands、brain and silk gland. The two results has an obvious consistency.These results laid a good foundation of further studies of Bm-P312.According to the experiment, we audaciously presumed that Bm-p312 may play some role in the reproductive development process of Bombyx mori. But the function of Bm-p312 should be demonstrated by further study.
