水体Cu2+胁迫对克氏原螯虾(Procambarus clarkii)体液免疫功能的影响
Effect of copper on the humoral immunity of Procambarus clarkii
为研究水体中不同浓度的Cu2+对克氏原螯虾体液免疫功能的影响,分别采用0(对照)、0.5、1.0、3.0、5.0、10.0 mg/L Cu2+刺激克氏原螯虾,24 h、48 h、72 h和96 h后测定其血清的抗菌活力Ua、溶菌活力 Ub、酚氧化酶活力PO以及超氧化物歧化酶SOD、过氧化氢酶CAT、丙二醛MDA的活性。结果表明,与对照组相比,1.0 、3.0、 5.0和 10.0 mg/L组分别在染毒72h和48h后,血清Ua活力和Ub活力显著下降(P ≤ 0.05);PO活性的变化存在时间-剂量的效应关系,96h后各处理组均显著降低(P ≤ 0.05);3.0、5.0和 10.0mg/L组的SOD和CAT活力,与对照组相比在72h后均呈现出显著的抑制状态;丙二醛(MDA)含量随胁迫时间的延长而不断增加,水体Cu2+浓度越高,MDA在体内累积量越大。结果提示,水环境中重金属Cu2+对克氏原螯虾的体液免疫因子活性具有显著的抑制效应,对机体健康状况构成潜在的威胁。
In order to study the effects of copper on the humoral immunity of crayfish Procambarus clarkii,crayfish were exposed to different concentrations of copper at 0, 0.5, 1.0, 3.0, 5.0, 10.0 mg/L. Antibacterial activity (Ua), bacteriolytic activity (Ub), phenol oxidase (PO), superoxide dismutase (SOD), catalase (CAT) and malondialdehyde (MDA) were determined after 24 h, 48 h, 72 h and 96 h of exposure, respectively.The results showed that the activities of Ua and Ub were lower than that in the control group (P ≤ 0.05) after 48h and 72h, when crayfish were exposed to the concentrations of 1.0, 3.0, 5.0 and 10.0 mg/L. PO activity reflected the time-dose response relationship, and PO of each treatment group was significantly lower than that of the control after 96h. Activities of SOD and CAT in 3.0, 5.0 and 10.0 mg/L groups was inhibited significantly during entire experimental period, but MDA in each tested group was higher than that of the control after 48h of exposure (P ≤ 0.05). The results suggested that Cu2 + in water had a significant negative effect on the humoral immunity of Procambarus clarkii, which indicated that heave metal pollution had potential threat to the health of crayfish.
