应用PCR-DGGE技术分析AO工艺中微生物多样性

为了了解AO工艺在处理城市污水过程中活性污泥的微生物群落多样性以及不同的生物处理池中微生物群落的相似性,从南京江心洲污水处理厂的好氧池和厌氧池采集活性污泥样品。采用基于SDS的细胞裂解法提取了污泥样品的DNA,获得基因组DNA大小达到了23Kb,采用PCR-DGGE技术对提取出的DNA进一步分析。结果表明:2个不同的生物处理池的微生物的种类都相当丰富,好氧池的细菌全部存在于厌氧池中;采用Quantity One分析软件计算的二者的相似性系数达到了75.2%。实验结果向污水处理厂的处理策略提供更多的理论依据

o find out the microbial diversity of actived sludges and the similarity of microbial community in different biological contact tanks, activated sludges in the aerobic tank and anaerobic tank of JiangXinZhou wastewater treatment plant in Nanjing city were collected. Cell cracking method based on SDS was used to extract the DNA in the activated sludges, then genomic DNA were acquired about 23 kilobase. Further analysis of DNA extracted was performed by PCR-DGGE.The result is described as follows: microbial species in two kinds of biological contact tanks are quite abundant,all microorganisms in the aerobic tank exists in the anaerobic tank.The similarity coefficient calculated by software Quantity One reaches 75.2%. More theoretical bisis is provided for prcessing strategy of wastewater treatment plants from the paper.
