虫草多糖对单侧输尿管梗阻小鼠肾小管上皮间充质转化的影响及机制探讨
Effect of Cordyceps polysaccharide on the tubular epithelial mesenchymal transition in mice unilateral ureteral obstructive
摘 要:目的 探讨虫草多糖对单侧输尿管梗阻(unilateral ureteral obstruction, UUO) 小鼠肾小管上皮-间充质转化的影响。 方法 建立小鼠UUO模型前2天开始,治疗组每天给予160mg/kg 虫草多糖灌胃。手术组和对照组均给与等量的蒸馏水灌胃。观察模型在第7天时肾间质纤维化程度及肾小管α-平滑肌肌动蛋白(α-SMA)、肾小管上皮表型标记物(E-cadherin)、结缔组织生长因子(CTGF)、整合素连接激酶(ILK)的表达分布。 结果 虫草多糖可以显著减轻肾间质纤维化(P<0.01)。 治疗组的α-SMA、CTGF、ILK蛋白的表达显著低于对照组(P<0.01)。而E-cadherin的表达显著高于对照组(P<0.01)。 结论 虫草多糖可显著抑制CTGF、ILK的表达,并抑制E-cadherin的丢失和 α-SMA的表达,提示其对肾小管间质纤维化的形成具有抑制作用。
Objective: To investigate the effect of cordyceps polysaccharide on the tubular epithelial mesenchymal transition in unilateral ureteral obstructive mice. Methods: CD1 Mice were randomly divided into three groups:control group (group C), unilateral ureteral obstrucion group (group UUO) and UUO treated with cordyceps polysaccharide (group UUO+Cp). Mice subjected to UUO were treated with Cp (160 mg/Kg )or vehicle respectively 2 days before the operation. Mice were sacrificed at day 7 after UUO operation. The expression of a-smooth muscle actin (a-SMA)、E-cadherin、connective tissue growth factor (CTGF)、integrin linked kinase (ILK) in kidney was assessed by immunohistochemistry and real time PCR. Results: Cp treatment significantly reduced intestitial fibrosis. The expression of a-SMA protein in the treatment group were significantly reduced compared to UUO group. The expression of E-cadherin protein in the treatment group were distinctively higher than those in the UUO group (P<0.01). There was a weak expression of CTGF and ILK in normal kidney, while a marked increasing expression of these markers was found in UUO group (P< 0.01). The expression of CTGF and ILK protein in the treatment group were distinctively inhibited compared to UUO group (P<0.01). Conclusion: Cp could significantly inhibit the over expression of CTGF and ILK in obstructed kidney, suppressing the loss of E-cadherin and the expression of α-SMA, which indicated that Cp could play an inhibitive role in the progression of renal tubulointerstitial fibrosis.
