电针对局灶性脑缺血再灌注大鼠迟发性细胞凋亡的影响
Effects of electroacupuncture against late-occurred neuron apoptosis of rat with focal cerebral ischemia/reperfusion
目的:观察电针对局灶性脑缺血再灌注大鼠缺血区神经细胞凋亡的保护作用及机制。方法:60只清洁级雄性SD大鼠随机分为假手术组、模型组、电针组,每组20只,模型组与电针组大鼠采用线栓法栓塞大脑中动脉60 min后拔出线栓恢复血流,电针组在缺血再灌注2 h后开始电针刺激(百会、水沟、足三里,疏密波,频率2Hz/15Hz)30 min,每隔12 h重复刺激1次。再灌注72 h后,TUNEL法观察电针对缺血区神经细胞凋亡的影响,免疫印迹法检测电针对缺血区神经细胞凋亡蛋白Bax、Bcl-2的影响。结果:电针组大鼠脑组织神经细胞凋亡数目较模型组明显减少(P<0.01),促凋亡蛋白Bax表达降低、抗凋亡蛋白Bcl-2表达增高。结论:电针刺激可明显减少脑缺血再灌注大鼠脑组织神经细胞凋亡数量,通过调节Bax/Bcl-2的比值来实现对缺血性神经元的保护作用。
Objective: To observe the effects of electroacupuncture(EA) against apoptosis in rat with focal cerebral ischemia/reperfusion (IR) injury and its mechanism. Methods: Sixty male SD rats of clean grade were randomly divided into sham operation group, model group and EA group (n = 20), Model group and EA group rats were modified the middle cerebral artery occlusion(MCAO) models with nylon monofilament, the filament was left in place for 60 min and then withdrawn to let the blood reperfuse. EA group was stimulated with EA (at the acupoints of Baihui, Shuigou, Zusanli, sparse and dense wave alternately, frequence of 2HZ/15HZ) for 30 min, repeated the stimulation every other 12 h. After 72 h of reperfusion, TUNEL staining was performed to observe the effect of EA against neuron apoptosis after IR, Western blotting was performed to observe the effect of EA against the apoptins of Bax and Bcl-2. Results: Neuron apoptosis in brain tissue of the EA group significantly alleviated than the model group. The ratio of Bax/Bcl-2 was significantly reduced than the model group. Conclusion: The stimulation of EA can significantly reduce the number of apoptotic neurons in brain tissue, and the protective effect is realized by adjusting the rate of Bax/Bcl-2.
