目的 扩增EV71 SDLY01株全基因组序列,了解EV71分离株SDLY 01的基因组特征,为构建感染性cDNA克隆提供基础。方法 采集手足口病患者的粪便标本,将其接种于RD细胞中分离病毒。运用一步法RT-PCR技术分三段对病毒基因组进行全序列扩增,经拼接后运用DNAstar 和MEGA 4软件对序列进行分析。结果 SDLY 01基因组全长7408nt,具有EV71的基因组特征,5′UTR为 745nt,3′UTR为81nt,编码区为6582nt,编码一个2194aa的多聚蛋白。SDLY 01全基因组序列与C4亚型安徽阜阳株(EU703814)具有最高的核苷酸同源性为97.2%。在5′UTR区及结构蛋白P1区,SDLY 01与EV71各亚型的同源性均高于组外对照株。而在非结构蛋白P2、P3区及3′UTR区,SDLY 01与组外对照株G10的同源性高于EV71各亚型(除C4亚型)。对VP1区及3D区构建的系统进化树显示,SDLY 01均位于C4亚型的分支中。经序列比对,在3Dpol第263位出现了氨基酸的突变(I263V)。结论 该分离株为C4亚型,3Dpol第263位氨基酸的突变(I263V)可能与EV71的神经毒力有关。非结构基因在EV71的进化过程中产生了重要的作用。

Objective To amplify the genome sequences of an EV71 strain and understand the genome characteristics of SDLY 01, it will provide a basis for constructing an infectious cDNA clone. Methods Stool specimens were collected from a patient with hand, foot and mouth disease. The virus was isolated in RD cells. The full length genome was amplified in three sessions by one step RT-PCR, and the sequences were analyzed by using DNAstar and MEGA 4 software. Results The full length of SDLY 01 was 7408nt, contained 5′UTR 745nt, 3′UTR 81nt, and the coding region 6582nt, encoding a 2194aa polyprotein。The sequence aligements showed that SDLY 01 had the highest nucleotide homology with the Fuyang strain EU703814. It is 97.2%. In regions of 5′UTR and P1, the nucleotide identities between SDLY 01 and EV71 strains were higher than the control strain G10. However, the SDLY 01 nucleotide sequences exhibited the higher homology to G10 than EV71 strains except C4 subtypes in P2, P3 and 3′UTR regions. The phylogenetic analysis based on VP1 and 3D regions showed that SDLY 01 was located in the branch of C4 subtypes. There was an mutation I263V in 3Dpol by amino acid sequence analysis. Conclusion SDLY 01 belongs to C4 subtype. The mutation I263V in 3Dpol may be related to neurovirulence of EV71. The non-structural genes play an important role in the evolution of EV71.
