可可全基因组内含子和微卫星联合分析及应用

NA分子标记技术在植物遗传和育种研究中发挥了重要的作用。而基于PCR技术开发的分子标记,便于操作,高效经济,在当前应用较为广泛。针对单个类型的标记所存在的某些局限性,本研究利用生物信息学的方法,在可可基因组中联合开发了两种类型的标记----内含子多态性标记和SSR标记。通过对可可全基因组进行了大规模扫描,分别得到169,662个内含子区段和104,640 个SSR重复单元。基于上述两种多态性的区段,分别开发了137,622对IP标记和65,968对SSR标记。经电子PCR验证,100%的引物在可可基因组中得到扩增产物。IP标记和SSR标记在可可基因组中的联合应用,将为可可的进化、分类和遗传多样性研究提供参考。同时,也为IP标记和SSR标记的联合应用于其他动植物研究中,提供了良好的探索。?????
NA molecular markers play an important role in the genetics and breeding of plants. It was used widely for the molecular markers based on the PCR technology, as its easy operation, economical and efficient. But there had some limitations for a single markers using in practical application. With the methods of bioinformatics, two types of markers, IP and SSR markers were combined developed in the genome of Cacao. After being large-scaled scan in the genome, 169,662 IP fragments and 104,640 SSR units were obtained. Based those segments, a total of 137,622 IP markers and 65,968 SSR markers were developed at finally. After testing by e-PCR, the results showed that 100% of those markers could obtain the target products in the Cacao genome. With the combined use of IP and SSR markers in Cacao, it can be well promote the development of evolution, classification and diversity study. Further on, it will also be a good sample for those two markers combined using in other animals and plants.
