郭志强教授经验方对子宫内膜癌移植瘤TRL4/MyD88表达的影响

目的：通过检测郭志强教授经验方对裸鼠内膜癌皮下移植瘤组织中TLR4/MyD88介导的肿瘤相关巨噬细胞的影响，探讨郭志强教授经验方抗子宫内膜癌的作用机制。方法：收集对数生长期的人子宫内膜癌Ishikawa细胞接种于裸鼠皮下，建立裸小鼠的移植瘤模型。将22只荷瘤裸鼠随机分模型组，中药预防组，中药治疗组，西药组，空白对照组，分别处理。观察裸鼠生活情况并定期监测移植瘤体积，6周后处死取材，通过酶联免疫法（Elisa）、实时荧光定量聚合酶链反应（RT-PCR）检测血清及瘤体组织内TRL4及MyD88表达。结果：西药组及两中药组肿瘤生长速度相当，无显著性差异（P> 0.05），三组均明显低于空白对照组（P< 0.05）；西药组、中药预防组、中药治疗组较空白对照组TLR4/MyD88的蛋白表达明显降低，差异具有统计学意义（P< 0.05）；西药组、中药预防组、中药治疗组较空白对照组TLR4/MyD88的mRNA表达明显降低，差异具有统计学意义（P< 0.05）。结论：郭志强教授经验方在基因水平具有抗肿瘤的作用，其作用机制可能与TLR4/MyD88介导的肿瘤相关巨噬细胞有关。
Objective To study the influence of the empirical formula of professor Guo Zhiqiang to the human endometrial carcinoma in the nude mice and the mechanism by investigating the expression of tumor-associated macrophages(TAMs) that mediated by the signal path of TRL4/MyD88.Methods Establish nude mouse transplantation tumor model by Ishikawa cells, and gave them different treatment respectively. Measured the tumor on time and detected the tumor by Elisa and Pcr methods. Resaults The expression of TRL4 and MyD88 were significantly more in the control group than that in the other three groups (p<0.05), but there was no significantly diffidence between the progesterone group and the two groups that were given by decoction. Conclusions the empirical formula of professor Guo Zhiqiang really have antitumor effect at the genetic level, its mechanism may be associated with TLR4 / MyD88 mediated tumor associated macrophages.?????
