叶下珠提取物抑制急性乙肝小鼠模型中HBV复制及表达的实验研究
Extract from Phyllanthus urinaria L. inhibits HBV replication and expression in a murine model of acute hepatitis B virus infection
目的 观察叶下珠提取物在小鼠急性乙型肝炎病毒(HBV)感染模型中对HBV复制和表达的影响。 方法 尾静脉注射C57b/6小鼠1.3倍的HBV真核表达质粒pHBV1.3,20 ug/只,建立小鼠急性乙型肝炎感染模型;将叶下珠提取物分别配制成0.5 g/ ml,0.25 g/ ml,0.125 g/ ml的药液。感染后第1 d起,予0.5 g/ ml(高剂量),0.25 g/ ml(中剂量),0.125 g/ ml(低剂量)药物1 ml灌胃,连续给药3天;同时设置正常组(1 ml 无菌PBS溶液灌胃)及病毒组。感染后第4 d,化学发光法检测血清HBs Ag、HBe Ag水平,PCR检测血清HBV DNA含量;免疫组化法检测肝组织HBc Ag、HBs Ag表达。 结果 叶下珠提取物高、中、低剂量组均能明显降低感染小鼠血清中HBs Ag、HBe Ag含量,与病毒组相比有显著性差异(p<0.001),其作用具有剂量依赖性;高剂量组药物显著降低了血清中HBV DNA水平,与病毒组相比有显著性差异(p<0.001),中、低剂量组中血清HBV DNA含量降低,然而与病毒组相比无显著性差异(p>0.05);高剂量组药物明显抑制了肝组织了HBc Ag及HBs Ag的表达。 结论 叶下珠提取物明显抑制了小鼠急性乙型肝炎感染模型中HBV的复制与表达,具有直接抗病毒作用。
Objective To observe the effects of the extract from Phyllanthus urinaria L. on HBV replication and expression in a murine model of acute hepatitis B virus infection. Methods The eukaryotic expression plasmid pHBV1.3, which contains 1.3-fold-overlength genome of HBV, was transfected into C57b/6 mice by hydrodynamic injection via tail vein, to establish a murine model of acute hepatitis B virus infection. The extract from Phyllanthus urinaria L. was prepared in 0.5 g/ ml, 0.25 g/ ml, 0.125 g/ ml , respectively, in sterile PBS. From the 1st day after infection, the mice were treated by intragastric administration with Phyllanthus urinaria L. at different concentration for 3 days. On days 4 after infection, chemiluminescence method was used to detect the concentration of HBs Ag and HBe Ag in mice serum; PCR method was used to detect the level of HBV DNA in serum; |immunohistochemistry was used to detect the expression of HBc Ag and HBs Ag in mice liver. Results The concentration of HBs Ag and HBe Ag in mice sera were decreased obviously by the drug in a dose-dependent manner, compared to the infected mice (P<0.05). Although the drug at all concentration could reduce HBV DNA concentration in mice sera, only that in the highest dose robustly reduced HBV DNA level, compared to the infected mice (P<0.05). And the drug in 0.5 g/ ml concentration obviously inhibited the HBc Ag and HBs Ag expression in mice liver. Conclusion The extract from Phyllanthus urinaria L. obviously inhibited HBV replication and expression in a murine model of acute hepatitis B virus infection and exerted distinctive anti-HBV effects.?????
