β-防御素抑制巨噬细胞自噬促进铜绿假单胞菌清除
β-defensins inhibit autophagy to promote macrophage-mediated clearance of Pseudomonas areuginosa
目的:探究β-防御素(β-defensin, BD)在巨噬细胞抗铜绿假单胞菌(Pseudomonas areuginosa, PA)感染中的作用与机制。方法:使用β-防御素或雷帕霉素刺激刺激THP-1巨噬细胞,流式细胞术和细菌平板计数实验分别检测巨噬细胞对PA的吞噬和胞内杀伤;通过免疫荧光和Western Blot检测微管相关蛋白轻链3(LC3)斑点聚集和表达,从而检测巨噬细胞自噬。结果:β-防御素抑制巨噬细胞自噬,自噬抑制巨噬细胞对PA的吞噬和杀伤,且β-防御素通过抑制巨噬细胞自噬促进对PA的吞噬和胞内杀伤。结论:β-防御素通过抑制巨噬细胞自噬促进巨噬细胞对PA的清除。
Objective: To investigate the role and mechanism of β-defensins in macrophage-mediated immune defence of Pseudomonas areuginosa (PA). Methods: THP-1 macrophages were treated with BD2, BD3 or rapamycin, followed by PA infection. Flow cytometry and bacterial killing assay were used to detect macrophage-mediated phagocytosis and intracellular killing of PA. Protein levels of LC3 were detected by Western Blot and LC3 puncta were observed by immunefluorescence microscopy. Results: β-defensins inhibited macrophages autophagy and autophagy inhibited phagocytosis and bacterial killing of PA. β-defensins inhibited autophagy to promoted macrophages--mediated phagocytosis and intracellular killing of PA. Conclusions: β-defensins inhibited autophagy to promoted macrophages--mediated clearance of PA.
