天然磷酸酯模拟物的研究在开发酶抑制剂方面一直备受人们关注。尽管运用生物电子等排原理得到的α,α-二氟亚甲基膦酸酯在开链磷酸酯模拟方面取得了较大的进展,然而环磷酸酯模拟物α,α-二氟亚甲基膦内酯的合成方法以及生物活性研究至今鲜有报道。本文从α, α-二氟亚甲基-β-联烯膦酸单酯出发,考察了其在亲电试剂DMTSF参与下的分子内关环反应,在乙腈为溶剂,0oC的条件下高区域选择性合成了一种新型环磷酸酯模拟物-六元环状的γ-甲硫代的α, α-二氟亚甲基膦内酯,产率中等到良好。

Research of natural phosphate mimics has been attracted much attention for their important role in the development of potential enzyme inhibitors. Despite the use of biological isostere principle to get α,α-difluoromethylenephosphonates in acyclic phosphate mimics has made great progress, in contrast, synthetic methods and the biological activities of cyclic phosphate mimics-α,α-difluoromethylenephostones still remain unexplored. In this paper, from the studies on the DMTSF mediated intramolecular cyclization of α,α-difluoromethylene-β-allenic phosphonic acid monoesters,a novel cyclic phospate mimics-six-membered γ-SCH3-substituted α,α-difluoromethylenephostones were prepared in moderate to good yields with high regioselectivity under the condition CH3CN /00C.
