不同类群动物胰蛋白酶结构特征与酶学性质关系
Relating Enzymologic Properties with Structure Features of Trypsin from Differential Animal Groups
胰蛋白酶(EC 3.4.21.4)在动物消化道中主要起蛋白消化及酶原激活作用,且在昆虫、甲壳动物蜕皮以及增强免疫力过程中起着至关重要的作用。选取已报道的完整胰蛋白酶序列,比较昆虫(I-TRY)、温热带甲壳动物(TTC-TRY)、冷水性鱼类(CAF-TRY)、温热带鱼类(TTF-TRY)和高等脊椎动物(HV-TRY)胰蛋白酶结构特征以及酶学性质差异。结果表明,作为无脊椎动物和脊索动物的进化地位最高的两个类群,节肢动物和脊椎动物胰蛋白酶氨基酸数目、分子直径、结构多样性、酶功能多样性进化趋势存在差别。动物胰蛋白酶活性部位及附近大多数氨基酸侧链严格保守的,由于外周和活性中心部位附近精氨酸、谷氨酸、天冬氨酸、组氨酸、半胱氨酸、丙氨酸、脯氨酸、酪氨酸和甲硫氨酸的不同,不同类群动物胰蛋白酶稳定性、柔性和反应耗能量差异显著,CAF-TRY、TTF-TRY和TTC-TRY催化常数(Kcat)升高或米氏常数(Km)降低,催化效率CAF-TRY>TTF-TRY≥TTC-TRY ≥I-TRY、HV-TRY。
rypsin (EC 3.4.21.4) is one of the major proteinases in animal digestive system and acts as a key mediator during the food assimilation and zymogen activition of animals, and exuviation and immunity improving of insects and crustuceans. 112 sequences obtained from Genebank were divided into five groups, and relationship between the structure features and enzymologic properties of trypsins from insects, temperate and tropical zone crustacean, cold adapted fish, temperate and tropical zone fish and higher vertebrate are conducted. It was indicated that, the evolutionary trend between the nummbers of amino acids, molecular diameter, diversity of molecular structure and fuction of arthropods and vertebrates trypsins are different, as two groups with highest evolutionary position within invertebrates and chordates separately. The animal trypsin active centre and amino acid side chains round are strictly conservative. With different Arg, Glu, Asp, His, Cys, Pro, Tyr and Met around periphery and active centre, significant difference of stability, flexibility and peaction energy consumption are present. The Kcat of CAF-TRY, TTF-TRY and TTC-TRY are increased, or the Km of which are reduced, hence the CAF-TRY catalytic efficiency > TTF-TRY≥TTC-TRY ≥I-TRY and HV-TRY.
