FSH和17α,20β-DHP对斑马鱼卵母细胞MAPK的作用及MAPK的表达定位
he Effects of FSH and 17α,20β-DHP on the MAPK activity and the Localization of MAPK During the First Meiosis in ZebraFish Oocytes
用不同浓度的FSH和17α,20β-DHP分别作用于斑马鱼卵母细胞,结果表明,1IU/ml的FSH和5ng/ml的17α,20β-DHP对斑马鱼卵母细胞GVBD都具有明显促进作用。其中17α,20β-DHP的作用效果较好,经过SDS-PDGE电泳和Western Blot检测发现,5ng/ml 17α,20β-DHP作用培养的斑马鱼卵母细胞后,MAPK被激活,卵母细胞的GVBD增多。免疫组化检测证实,发育第I期的卵母细胞中MAPK没有激活,第II期、第III期细胞质中MAPK开始激活,第V期卵母细胞细胞质与核区同时存在有活性的MAPK,并伴有GVBD发生。以上结果说明MAPK在斑马鱼卵母细胞第一次减数分裂恢复过程中发挥重要作用,并且随着MAPK激活,其活性部位会发生从质到核的转移。
Zebrafish oocyte maturation were induced by various concentrations of FSH and 17α,20β-DHP.The results indicated that both of FSH and 17α,20β-DHP initiated germinal vesicle breakdown (GVBD). The 5 ng/ml of 17α,20β-DHP significantly increased GVBD of oocytes and acitivated MAPK. The GVBD increased with the MAPK activity enhance induced by 17α,20β-DHP. The histochemistry assay showed that MAPK was activated at the oocyte development stages II and III in the oocyte plasm while no MAPK activity was observed at the stage I. At the stage V, the acitvated MAPK could be observed both in the plasm and nucleus .The results indicated that activated MAPK played an important role in promoting the ovary development.
