小鼠CLOCK基因对食欲素orexin的调节作用
Regulation of orexin by influence clock in mice
在生物的近日节律系统中,clock基因起着核心作用。近年的研究发现,clock和肥胖、发育和睡眠有着重要联系,这与之前发现的同样对肥胖和睡眠具有调节作用的食欲素(orexin)似乎扮演了相似的角色。本实验室在前期的研究中发现,小鼠下丘脑orexin的表达也具有明显的近日节律。为了探究clock和orexin之间是否存在相互调节作用,我们向小鼠侧脑室注射干扰表达clock的siRNA,分别用Real-time PCR技术测定Orexin的mRNA表达情况和用Western-blot技术测定Orexin蛋白的表达情况,发现实验组和对照组间并无统计学差异。为了更进一步确定clock在调节orexin功能中的作用,本实验又采用了orexin和c-fos免疫荧光双染,发现clock被成功干扰后orexin阳性神经元表达活性显著降低。
lock gene plays a central role in circadian system. In recent years, the study found that clock gene also has relationship with obesity and sleep duration. It seems plays the same role with orexin whose regulation function in obesity and sleep has been proved before. In previous research, we find that orexin which express in hypothalamus has its own circadian rhythm in mice. In order to explore whether exists regulation between clock and orexin, we inject clock siRNA to mice paracele, measuring the mRNA and protein expression level of orexin with Real-time PCR and western-blot, finding that there is no statistical differences between experimental group and controlled group. To further determine the clock role in regulating function of orexin, we use immunofluorescent double staining with orexin and c-fos, the result shows that orexin positive neurons express activity decreased significantly when clock been influenced successfully.
