目的 探究G1(GPER 特异性激活剂)和双酚A(BPA)对血管内皮细胞窖蛋白-1(Cav-1)以及Cav-1和G蛋白偶联雌激素受体(GPER)相互作用的影响。方法 不同剂量的双酚A或G1干预下,Western Blot方法检测内皮细胞中Cav-1、HSP90蛋白表达水平和PKA磷酸化水平和免疫荧光检测Cav-1和GPER之间的相互作用。结果 高浓度的双酚A或G1都能上调内皮细胞Cav-1蛋白的表达;其中G1可促进Cav-1和GPER的相互作用,双酚A可降低HSP90蛋白表达水平并上调PKA信号蛋白的磷酸化水平。结论 1. GPER作用的发挥与Cav-1密切有关;2. 双酚A影响血管内皮细胞Cav-1的表达调控以及HSP90的表达水平,并可能通过PKA信号通路促进内皮的增殖和迁移。

To explore the effects of G1 and bisphenol A on cavelin-1 in vascular endothelial cells and the interaction between Cavelin-1 (Cav-1) and G protein-coupled estrogen receptor (GPER). Methods: After different doses of BPA or G1 for 24 h treatment, expression levels of Cav-1, HSP90, and PKA phosphorylate substrate was measured by Western Blot. The interaction between Cav-1 and GPER under the treatment of G1 was detected by immunofluorescence. Result: High concentration of bisphenol A and G1 up-regulated the expression of Cav-1. G1 stimulated the interaction between the GPER and Cav-1, while the phosphorylation levels of PKA and the expression of HSP90 were elevated by bisphenol A. Conclusion: 1.The function of GPERB exhibits close connection to Cav-1. 2. Bisphenol A, which possibly results in proliferation as well as migration in endothelial cells through downstream proteins PKA on Cardiovascular System, also affects the expression of caveolae and HSP90.
