新型蛋白标记荧光探针的合成及生物学测试

在甲基磺酸作用下,以2,4-二氯间苯二酚和3,6-二氯-4-羧基邻苯二甲酸酐为原料缩合得荧光素母体。该荧光素母体依次和三甲基乙酸酐、二异丙基胺盐、盐酸、三氟乙酸N-琥珀酰亚胺酯反应得到二新戊酸酯保护的荧光素N-琥珀酰亚胺酯。用6-氨基己酸和荧光素N-琥珀酰亚胺酯反应,然后用氨水脱去新戊酸酯保护基得到一种新型蛋白标记荧光探针。目标产物及所有中间体通过IR,1HNMR,MS和元素分析表征。对目标产物进行细胞标记生物学测试,并用共聚焦激光扫描显微镜扫描细胞图像,图像显示该荧光探针具有很好的荧光性、光稳定性和生物相容性。

novel chlorinated fluorescent dye was synthesized by the reaction of 2,4-dichlororesorcinol with 3,5-dichloro-2,4-dihydroxybenzonic acid catalyzed with methanesulfonic acid . The fluorescent was treated with trimethylacetic anhydride, diisopropylamine , HCl and N-hydroxysuccinimidyl trifluoroacetate to obtain a dipivaloyl-5(6)-carboxyfluorescein N-hydroxysuccinimidly ester. A fluorescent probe dye with space linker arm was obtained by the reaction of dipivaloyl-5(6)-carboxyfluorescein N-hydroxysuccinimidly ester with 6-aminoncaroic acid. The structures of target compound and intermediates were determined via IR, MS, 1HNMR and element analysis. The investigation in immunofluorencence histochemistry showed it has strong fluorescence, high photostability and good biocompatibility.
