茉莉酸甲酯调控阳春砂HMGR、DXR和DXS基因表达

【目的】探讨茉莉酸甲酯(Methyl Jasmonate,MeJA)调控阳春砂萜类化合物生物合成途径3个关键酶基因HMGR、DXR、DXS表达的规律。【方法】用20 μmol/L 和200 μmol/L两个浓度的MeJA喷施阳春砂幼苗,诱导不同时间后取样,提取RNA,荧光定量PCR法测定AvHMGR、AvDXR、AvDXS相对表达量。【结果】MeJA对AvHMGR、AvDXR和AvDXS的表达均有促进作用,其中对AvDXR的促进作用最明显,AvHMGR次之,两者最高表达量分别达到处理前的9.40和3.95倍;AvDXR基因受高低浓度MeJA的诱导表达均十分明显,而AvHMGR基因仅对低浓度MeJA敏感,AvDXS基因对高低浓度的MeJA均不敏感;AvHMGR和AvDXR的表达量在诱导后24 h达到最高峰,在72 h恢复到处理前水平。【结论】诱导子MeJA对AvDXR和AvHMGR基因表达的调控是激发式的,基因表达迅速上调到一定水平,而后逐渐回落。研究结果为利用MeJA及AvHMGR、AvDXR基因进行阳春砂萜类化合物的代谢工程调控提供依据。
Objective To investigate the rule of the MeJA regulating the expression of HMGR, DXR and DXS, which are the genes encoding the key enzymes of terpenoids biosynthetic pathway in Amomum villosum Lour. Methods The seedlings of A. villosum were sprayed with 20 μmol/L and 200 μmol/L MeJA. RNA was extracted from the leaves collected at different time after MeJA inducing. The expression levels of AvHMGR, AvDXR and AvDXS genes were detected by Real-time quantitative PCR. Results The expression of AvHMGR, AvDXR and AvDXS could be up-regulated by the induction of MeJA. The stimulation of AvDXR was most significant and that of AvHMGR was less significant. The highest expression levels of AvDXR, AvHMGR were 9.40 and 3.95 times as high as the untreated sample respectively. AvDXR was induced significantly by both high and low concentration MeJA, however, AvHMGR was only sensitive to the induction of MeJA with low concentration and AvDXS was insensitive to the induction of either high or low concentration MeJA. The expression of AvHMGR and AvDXR reached the highest level at 24 h and then returned to the former level at 72 h after the induction. Conclusion The regulation of elicitor MeJA inducing AvDXR and AvHMGR expression is excitation formula. The expressional levels of the genes reached the highest level rapidly and then fell back to the former level. This study provided the theory basis for using MeJA and AvHMGR, AvDXR to process the regulation of terpenoid metablic engineering for A. villosum.
