PTSD大鼠杏仁核神经元凋亡及TMP酶变化的研究
EXPRESSION OF TMPASE AND APOPTOSIS IN AMYGDALA NEURONS OF PTSD
目的:研究创伤后应激障碍(posttraumatic stress disorder,PTSD)大鼠杏仁核神经元细胞凋亡及三偏磷酸酶(Thiamine Monophosphatase TMP)活性的变化。方法:成年健康雄性Wister大鼠50只,随机分为SPS模型的1d、4d、7d、14d组及正常对照组。应用Annexin V-FITC/ PI双标记流式细胞技术分析PTSD大鼠杏仁核神经元细胞凋亡及定量检测;利用透射电镜观察杏仁核神经元超微结构改变;利用酶组织化学方法检测杏仁核神经元TMP酶活性的表达变化。结果:SPS刺激后:杏仁核神经元细胞凋亡率显著增加,TMP酶活性明显比正常对照组增强,均于第4d达高峰。7d和14d凋亡细胞逐渐减少、TMP酶活性减弱。结论:PTSD杏仁核神经元发生细胞凋亡,凋亡率增加的同时TMP酶活性相应增强,提示 TMP酶参与PTSD大鼠杏仁核神经元凋亡产物的吞噬。
Objective Study on apoptosis and Thiamine Monophosphatase (TMP) expression of amygdala neurons in rats of PTSD. Methods A total of 50 male Wistar rats were randomly divided into 1 d, 4 d, 7 d, 14 d groups of SPS and normal control group. Apply Annexin V-FITC/ PI double-labeled flow cytometry to analysis PTSD rats neurons in amygdala; Using transmission electron microscopy observation of neurons in the amygdala ultrastructural changes; Using enzymes to detect neurons in amygdala activity of TMP expression. Results Amygdala neuronal apoptosis was significantly increased after SPS, TMP activity was significantly enhanced more than the normal control group ,both of reached the peak at 4d and graddually decreased at 7d and 14d. Conclusion Apoptosis happened in amygdala neurons of PTSD rats, the apoptosis ratio increasing is associated with the enhancement of TMP activity in SPS model. This suggest that the TMP is involved in neuronal apoptosis products phagocytosis in amygdala.
