HIF羟脯化酶在大鼠卵巢细胞中在表达及其定位
HIF Prolyl Hydroylase Expression and Localization in Ovarian Cells of Rats
目的:利用成年SD大鼠卵巢卵泡发育模型,研究三种HIF羟脯化酶(HIF prolyl hydroylase, PHD)同工酶在卵巢细胞的表达及其定位。方法:选取20只3-4月龄的成年SD雌性大鼠,通过阴道涂片鉴定其发情周期,并通过免疫组化PowerVisionTM(PV)二步法观察PHDs在卵巢卵泡发育周期中的表达及其定位。结果:PHD1、PHD2、PHD3在卵母细胞、颗粒细胞、黄体细胞以及间质细胞中均有明显表达,但PHD2的表达量明显超过了PHD1和PHD3,并且在不同的卵巢卵泡发育阶段呈现规律性变化。结论:PHDs可能直接参与卵巢卵泡发育等卵巢生理功能的调节。
im: Using the model of ovarian follicle development of adult female SD rats, we investigated the expression and localization of three isozymes of HIF prolyl hydroylases (PHD1, 2and, 3) in ovarian cells. Methods: 3-4 months old SD female rats were selected to identify their estrous cycle by vaginal smears, and to observe the expression and localization of PHD proteins in ovarian follicular development cycle by two-step immunohistochemistry (IHC) PowerVisionTM (PV). Results: PHD1, PHD2 and PHD3 are all significantly expressed in oocyte, granulosa cells, corpus luteum cells and stromal cells; but the expression of PHD2 significantly more than the amount of PHD1 and PHD3; and their expression changed in stage-specific manner during the follicular developmental cycle in the ovary. Conclusion: PHDs may be directly involved in the regulation of ovarian physiological functions such as ovarian follicles development.
