不同频率电针对抑郁症模型大鼠行为学和β-内啡肽的差异性研究

目的]观察不同频率电针对抑郁症模型大鼠行为学和β-内啡肽的差异。[方法]制作慢性应激型抑郁症模型大鼠,分别予以氟西汀、假针、2HZ电针、100HZ电针治疗7天,测定行为学表现和β-内啡肽水平。[结果] 2Hz电针组和100Hz电针组与模型组、假针组、氟西汀组比较,糖水消耗量和水平运动、垂直运动都有差异(P<0.05),而氟西汀组与模型组、假针组并无差异。2HZ电针组和100HZ电针组比较,糖水消耗量和水平运动无差异,但垂直运动有差异(P<0.05)。2Hz电针和100Hz电针脑内β-内啡肽(β-EP)水平均降低(P<0.05),2Hz电针降低显著(P<0.05)。2Hz电针和100Hz电针β-EP水平均与糖水消耗量和水平运动有相关性,但2Hz电针组在纹状体表现出相关性,100Hz电针组在中缝核表现出相关性。[结论] 2Hz电针和100Hz电针对抑郁症模型大鼠均表现出抗抑郁快速起效特点,2Hz电针效果优于100Hz电针,该效应特点与β-EP水平降低有关。2种频率电针β-EP水平与行为学的相关性表现在脑的不同部位。β-EP的受体-μ-受体及其信号通路的研究将是进一步深入研究的目标。
Objective]To observe the behavioral performance and beta-endorphin levels of depression model rats treated with different frequencies of electro-acupuncture.[Methods] we made the chronic stress-induced depression model rats, which were treated with fluoxetine, sham acupuncture, 2HZ EA and 100HZ EA for seven days respectively; and measured the behavioral performance and beta-endorphin levels of rats. [Results] 2Hz EA group and 100Hz EA group both increased in sugar consumption, horizontal movement and vertical movement comparing with the model group, sham acupuncture group, the fluoxetine group respectively (P<0.05), while there was no difference among fluoxetine group, model group and sham acupuncture group. 2HZ EA group corresponds to 100HZ EA group in sugar consumption and horizontal movement, but 2HZ EA had an advantage in vertical motion (P<0.05).β-endorphin (beta-EP) decreased in 2Hz EA group and 100Hz EA group (P<0.05), and significantly in 2Hz EA group (P <0.05). 2Hz EA and 100Hz EA were both related to beta-EP level, sugar consumption and horizontal movement respectively, but 2Hz EA group shows in the striatum, and 100Hz EA group in the raphe nuclei. [Conclusion] 2Hz EA and 100Hz EA both show antidepressant rapid onset characteristics on depression model of rats, and 2Hz EA is more effective than 100Hz EA. This is related to the decreasing level of beta-EP. The correspondence between beta-EP and behavior performance showed in different parts of the brain caused by the double electro-acupuncture. μ-receptor, the receptor of beta-EP, and its signaling pathway is a further objective of the study.
