目的 研究三邻甲苯磷酸酯(TOCP)诱导的迟发性神经病(OPIDN)中自噬相关蛋白Atg1、Atg5和Beclin1的变化,探讨OPIDN发生的分子机制。 方法 成年罗曼母鸡30只,随机分为对照组、1d、5d、10d 和21d组(每组6只),除对照组外,其余各组动物均按750 mg/kg经灌胃一次性染毒TOCP,分别在相应时间点处死动物取胫神经和脊髓,免疫印迹法检测胫神经和脊髓中Atg1、Atg5和Beclin1的相对含量。 结果 TOCP染毒显著影响了母鸡神经组织中自噬相关蛋白Atg1、Atg5和Beclin1的表达。与对照组相比,胫神经中Atg1含量在TOCP染毒后1、5、10、21天分别下降了29.8%、64.4%、43.5%和19.8%(P<0.05);Atg5含量在TOCP染毒后1、5、10、21天分别下降了36.8%、49.6%、51.2%和31.5%(P<0.05);beclin1含量在TOCP染毒后1、5、10天分别下降了68.5%、66.3%和32.2%(P<0.05)。与对照组相比,脊髓组织中Atg1含量在TOCP染毒后1、5、10天分别下降了23.5%、48.7%和20%(P<0.05);Atg5含量在TOCP染毒后1、5、10、21天分别下降了32.7%、51.5%、47.3%和39.6%(P<0.05);beclin1含量在TOCP染毒后1、5、10、21天分别下降了28.9%、50.2%、43.2%和28.3%(P<0.05)。结论 TOCP染毒干扰了母鸡神经组织中自噬相关蛋白的表达,这可能与OPIDN的发病机制有关。

Objective To study the changes of autophagy-related proteins in the delayed neuropathy (OPIDN) induced by hree-o-cresyl phosphate (TOCP), and explore the molecular mechanism of OPIDN. Methods Thirty adult Roman hens were randomly divided into groups of control, 1d, 5d, 10d and 21d(6 hens per group). Except the group control,the rest were intoxicated with TOCP by gavage at a single dosage of 750 mg/kg. The level of Atg1, Atg5 and Beclin1 in tibial nerves and spinal cords was determined by immunoblotting. Results In tibial nerves, compared to the control group, the level of Atg1 decreased by 29.8%, 64.4%, 43.5% and 19.8% respectively on 1d, 5d, 10d and 21d(P<0.05); the level of Atg5 decreased by 36.8%, 49.6%, 51.2% and 31.5% respectively on 1d, 5d, 10d and 21d(P<0.05); the level of Beclin1 decreased by 68.5%, 66.3% and 32.2 respectively on 1d, 5d and 10d(P<0.05). In spinal cords , compared to the control group, the level of Atg1 decreased by 23.5%, 48.7% and 20% respectively on 1d, 5d and 10d(P<0.05); the level of Atg5 decreased by 32.7%, 51.5%,47.3% and 39.6% respectively on 1d , 5d, 10d and 21d(P<0.05); the level of Beclin1 decreased by 28.9%, 50.2%, 43.2% and 28.3% respectively on 1d, d5, 10d and 21d(P<0.05). Conclusion The intoxication of TOCP was associated with the significant change of autophagy-related proteins in hen nervous tissues, which might be involved in the pathogenesis of OPIDN.
