番茄果实成熟突变体Nr和Rin荧光定量PCR内参基因的选择

实时荧光定量PCR作为分析基因表达水平的常用方法, 因其具有的高灵敏性和高特异性,逐渐在功能基因组学研究中发挥着重要的作用。在分析功能基因的表达水平过程中,校正表达数据所选择的内参基因对结果可靠性起着关键的作用。以野生型番茄和两个果实成熟突变体Nr及Rin分别的五个果实发育时期的果实为研究对象, 采用实时荧光定量PCR 这一方法, 对三个候选的内参基因CAC、EXP和GAPDH的表达稳定性进行了评价, 经过数据分析程序geNorm 和NormFinder对以上结果的稳定性分析, 发现3个候选内参基因在突变体和野生型番茄的果实发育不同时期表达相对都较为稳定, 综合两种软件分析的结果发现表达最为稳定的是CAC, 表明该基因可以用作番茄突变体果实成熟过程中基因表达水平分析的内参基因。
s a commonly used method to analyze the level of gene expression with higher sensitivity and specificity, it is important to select an appropriate reference gene for normalization of experimental data when using real-time quantitative PCR for analysis. In this study, two statistical analysis program geNorm and NormFinder was applied for evaluating the gene expression stabilities of three housekeeping genes (CAC, GAPDH and EXP) in the different growth phases of tomato fruit. The results showed that the best reference gene was CAC which was the most stable gene and could be used for the normalization of experiments data by real-time quantitative PCR.?????
