背景:前期研究发现miRNA-126-3p可能通过调控VEGFA参与先天性心脏病相关性肺动脉高压发病,本研究拟通过动物模型进行验证。目的:探讨miRNA-126-3p与大鼠肺动脉高压的发病机制是否有关。方法:25只雄性SD大鼠,其中实验组(15只)采用野百合碱诱导,对照组10只,四周后,采用qRT-PCR检测其肺组织miRNA-126-3p的表达,并从mRNA水平和蛋白水平检测VEGFA。结果:野百合碱诱导组有8只造模成功,平均肺动脉压力较对照组有明显差异(p< 0.0001);实验组miRNA-126-3p表达水平较对照组有明显统计学差异(p< 0.0001);实验组的VEGFA表达在mRNA水平和蛋白水平都较对照有明显统计学差异(p< 0.0001)。结论:在野百合碱诱导的肺动脉高压大鼠中,miRNA-126-3p可能通过调控VEGFA参与肺动脉高压发生发展。?????

Background: Our previous studies have found that the miRNA-126-3p may participate in pulmonary hypertension (PH) of congenital heart disease by regulating VEGFA; this study intends to validate it by animal models. Objective: To explore whether miRNA-126-3p is related to the occurrence of PH in rats. Methods: The experimental group was (15 male SD rats) induced by monocrotaline, and control group had 10 rats. Four weeks later, the qRT-PCR detection of the lung tissue expression of miRNA-126-3p was done, and the level of mRNA and protein level detection of VEGFA also was tested. Results: There were 8 rats which were successfully induced by monocrotaline, and the average pulmonary artery pressure had obvious difference than the control group (p<0.0001); miRNA-126-3p expression level was significantly statistical difference (p<0.0001) between the two groups; VEGFA expression at mRNA level and protein level were statistically significantly than control group (p<0.0001). Conclusion: miRNA-126-3p may participate in monocrotaline induced PH in rats by regulating VEGFA.
