5-羟甲基胞嘧啶上调参与抑郁样行为的发生
Upregulation of 5-hmC involved in the occurrence of depressive-like behavior
目的:观察5-羟甲基胞嘧啶(5-hmC)在正常ICR小鼠脑组织中的表达分布以及抑郁发生后小鼠脑组织不同部位5-hmC的表达变化。方法:通过心脏灌注冰冻切片以及免疫组织化学技术检测正常小鼠脑组织5-hmC的表达分布;采用免疫荧光染色以及Dot blot观察线粒体5-hmC的表达;采用空间行为限制建立小鼠抑郁样表型模型,并通过悬尾实验(TST)和强迫游泳实验(FST)检测小鼠是否存在抑郁样表型;采用Dot blot方法以及免疫组织化学技术(IHC)检测抑郁后小鼠脑组织5-hmC的表达变化。结果:(1)DAB染色直观地展示了5-hmC在正常小鼠脑组织中不同部位的分布情况,其中大脑皮层、海马、下丘脑、杏仁核、纹状体、小脑以及脑干等部位5-hmC含量较为丰富;并且5-hmC在核基因和线粒体DNA中均有表达。(2)行为学实验结果表明,与对照组相比,实验组小鼠的悬尾实验和强迫游泳实验的不动时间均明显延长 (P<0.05)说明造模小鼠产生抑郁样表型;(3)Dot blot结果显示Stress组小鼠脑组织中5-hmC的含量明显增高 (P<0.05);免疫组织化学染色方法检测显示Stress组脑组织皮层、下丘脑、海马以及杏仁核部位5-hmC含量较对照组增高 (P<0.05)。
Objective: To observe the distribution of 5-hmC in mice brain and the change of 5-hmC in the depressive mice, then to explore the possible pathogenesis of depressive phenotype. Methods: Heart perfusion and frozen section technique were used to normal adult male ICR mice;Then we used immunohistochemical staining method to obeserve the distribution of 5-hmC in brain tissue. Immunofluorescent staining and Dot blot were used to obeserve the distribution of 5-hmC in mitochondria. Chronic restraint stress was used to establish mouse model of depression. Tail suspension test and forced swimming test were used to examine depressive phenotype in mice. Using Dot blot and IHC techniques to detect the contents of 5-hmC in the brain tissue.Results: (1) DAB staining of normal male ICR mice intuitively showed the distribution of 5-hmC in the brain tissue; 5-hmC expressed rich in the cerebral cortex , hippocampus , hypothalamus , amygdala , striatum , cerebellum and brain stem; In addition, it has expressed in nuclear genes and mitochondrial DNA. (2) Tail suspension test and forced swimming test showed that the immobility time of Stress group were significantly increased compared with the Control group (P<0.05).(3) Dot blot showed that the level of 5-hmC in brain tissue of stress group increased significantly (P<0.05). After the depression , Immunohistochemical staining showed that 5-hmC increased in the cerebral cortex , hypothalamus , hippocampus and amygdala(P<0.05).
