马身猪高肌内脂肪含量形成的机制研究

肌内脂肪含量(IMF)是影响猪肉品质的重要因素之一,其含量高低直接影响猪肉的多汁性与适口性。马身猪是优良的地方品种,以IMF含量高、肉质优良著称,但其高IMF形成机制尚不明确。本研究以1月龄和6月龄山西马身猪与大白猪为研究对象,旨在探究马身猪高IMF形成的机制。研究采用索氏抽提法测定背最长肌IMF含量,运用Real time PCR与Western blot对脂肪细胞分化的关键基因及蛋白表达进行分析。结果显示,马身猪的IMF含量极显著高于大白猪。马身猪中涉及脂肪细胞分化的关键基因,包括C/EBPα(P<0.05)、PPARγ(P<0.01)、Zfp423(P<0.05)和C/EBPβ(P<0.05)表达均高于大白猪,而SREBP1表达差异不明显(P>0.05)。Western blot结果显示,马身猪中PPARγ(P<0.05)、C/EBPα(P<0.001)与FABP4(P<0.001)的含量均极显著高于大白猪。试验结果表明马身猪成脂细胞分化潜能大于大白猪,可为进一步研究骨骼肌肌内脂肪形成的机制提供依据。
he intramuscular fat (IMF), or marbling, is one of major factors affecting pork quality, and its content can influence the palatability and tenderness of meat. The Mashen pigs is well known for their high IMF, but the mechanism remains unclear. In present study, the Mashen pigs and Large white pigs (30 d and 180 d after birth) were used to explore the possible mechanism of higher IMF in Mashen pigs. The IMF contents were determined by Soxhlet extractor method, and the candidate key genes involved in adipogenesis by both Real-time PCR and Western blot. The results showed that the Mashen pig possessed higher IMF content in skeletal muscle. Real-time PCR results indicated that mRNA expressions of C/EBPα (P < 0.05), PPARγ (P < 0.01), Zfp423 (P< 0.05) and C/EBPβ(P < 0.05)were higher in Mashen pigs compared with Large white pigs. Accordingly, the protein contents of PPARγ (P< 0.05), C/EBPα (P < 0.001) and FABP4 (P < 0.001) were higher in Mashen pigs. The data suggest that the higher IMF content in Mashen pigs may attributed to the higher adipogenesis potential, which can provide basic data for further exploring the IMF formation in skeletal muscle of pigs.
