新抑癌基因DLC2对乳腺癌细胞增殖、迁移和侵袭的影响
Effects of Tumor Suppressor DLC2 on Breast Cancer Cell Proliferation, Migration and Invasion
目的:探讨新抑癌基因DLC2对乳腺癌细胞增殖、迁移和侵袭能力的影响。方法:在人乳腺癌细胞株MDA-MB-468 和MDA-MB-231中通过shRNA慢病毒载体构建DLC2低表达稳定细胞株,通过划痕实验、Transwell 迁移实验、Transwell 侵袭实验、克隆形成实验、细胞黏附实验、流式细胞术等方法观察DLC2表达下调对乳腺癌细胞生物学行为的影响。结果:下调DLC2表达后乳腺癌细胞的克隆形成能力、迁移能力和侵袭能力均增强,细胞黏附能力减弱,进入细胞周期S期的细胞增多,但对乳腺癌细胞凋亡没有影响。结论:DLC2负调控乳腺癌增殖、迁移和侵袭,提示其可能在乳腺癌发生发展中起抑癌基因的作用。
In this paper, we discussed the effects of tumor suppressor DLC2 on breast cancer cell proliferation, migration and invasion. We constructed DLC2 low expression stable human breast cancer cell lines in MDA-MB-468 and MDA-MB-231. Then, we compared the abilities of cellular proliferation, cloning formation, migration, invasion between DLC2 knockdown cell lines and control cell lines. We also compared the differences of tumor cell distribution in cell cycle by flow cytometry, and apoptosis by DAPI staining. We found loss of DLC2 inhibited cell adhesion, promoted cloning formation, cell migration and invasion in breast cancer. Knocking down DLC2 in breast cell lines, more tumour cells entered into S phase, but no impact on cell apoptosis. These indicated DLC2 regulated cell proliferation, migration and invasion, and played a tumor suppressor role in breast cancer.
