重组鹅 β-防御素4蛋白的原核表达及其生物学特性分析
Recombinant expression and biological analysis of goose avian β-defensin 4
本实验利用RT-PCR方法从鹅体内扩增出1个新的鹅β-防御素: AvBD4,其包含171个碱基,编码56个氨基酸;经同源性分析,鹅AvBD4与鸡AvBD4同源性最高为80.9%;为研究新鹅β-防御素的生物学活性,采用His标签蛋白原核表达系统,将该基因亚克隆到pProEX HTa上,构建出重组表达质粒。将重组表达质粒转化到大肠杆菌Rosseta 感受态中,用IPTG进行诱导,诱导后经Tricine-SDS-PAGE电泳分析,诱导表达的蛋白大部分以包涵体形式存在。重组蛋白抗菌活性测定结果表明,重组鹅AvBD4蛋白对大肠杆菌,鸡白痢沙门氏菌,四联球菌,枯草芽孢杆菌及金黄色葡萄球菌均具有抗菌活性(P<0.05),且随蛋白浓度的增加抑菌活性增强。此外,高浓度盐离子抑制重组蛋白的抗菌活性(P<0.05)。重组蛋白对鸡红细胞没有溶血活性(P>0.05)。
In this paper a novel goose β-defensins 4 was isolated from goose by RT-PCR. Sequence analysis showed the gene fragment of goose AvBD4 consisted of 171 bp encoding 56 amino acids. In addition, phylogenetic relationships between the goose AvBD4 genes from other avian species, and some mammalian beta-defensin were analyzed. It was demonstrated that goose AvBD4 shared high amino acid homology 80.9% with chicken AvBD4.The cDNA of goose AvBD4 were cloned into EcoR I and Sal I sites of pProEX HTa vector,The recombinant expression plasmids were translated into Ecoli Rosseta and the bacteria were expression induced by IPTG.It was demonstrated by Tricine-SDS-PAGE that the recombinant protein was produced as insoluble bodies in the cells. Additionally, the goose AvBD4 recombinant protein was purified. Micrococcus tetragenus, Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Salmonella pullorum were used to research the antibacterial activities of AvBD4 by colony counting method. The results indicated that the recombinant protein have different degrees of antibacterial activity. Furthermore, the effect of salt concentration on the antibacterial activity and hemolytic activity of the recombinant protein was investigated. The results indicated that in high salt ions conditions, antibacterial activity of recombinant protein was significantly decreased. The goose AvBD4 was not found significant hemolytic activity.
