辣椒素促进食管鳞癌细胞凋亡的体外研究

目的:本研究将检测食管鳞癌细胞EC9706和Eca109中NF-κB信号通路的状态;并探讨辣椒素是否能够通过阻断NF-κB信号通路对食管鳞癌细胞增殖、抗凋亡产生影响。方法:Western blot法检测EC9706和Eca109细胞质中IκBα及其磷酸化状态及不同浓度的辣椒素作用后pIκBα和抗凋亡蛋白Bcl-2的表达,p50和p65在细胞核中的表达;凝胶滞留实验(electrophoretic mobility gel shift assay,EMSA)检测细胞核中NF-κB与DNA的结合活性及辣椒素在不同浓度作用下对NF-κB与DNA结合活性的影响。在EC9706和Eca109细胞中加入辣椒素培养48h,流式细胞仪检测凋亡细胞情况。结果:Western blot的结果显示,IκBα和pIκBα在EC9706和Eca109的细胞质中均表达,p50和p65在细胞核中也表达;在辣椒素作用下EC9706和Eca109细胞中pIκBα和Bcl-2蛋白的表达水平逐渐下降,NF-κB与DNA的结合活性也逐渐下降。EC9706和Eca109在单独使用辣椒素后,就可促进细胞的凋亡(P<0.05);当与5-FU 联合使用时,可明显提高EC9706和Eca109细胞对化疗药的敏感性,凋亡细胞显著增加(P<0.05)。结论:辣椒素可以通过抑制IκBα的磷酸化阻断NF-κB信号通路,促进细胞凋亡。

OBJECTIVE: This study was to detect the status of NF-κB signaling pathway in two ESCC cell lines and evaluate whether capsaicin induced apoptosis through inhibition of NF-κB signaling pathway in ESCC cells. METHODS: Western blot was used to determine the protein levels of inhibitor-κBα (IκBα) and phosphorylated-IκBα (pIκBα), p50 and p65 in the nuclear extracts. Cytoplasmic extracts were examined for the expression of pIκBα and Bcl-2 using Western blot after the cells were treated with different concentration capsaicin. DNA-binding activity and the change of DNA-binding activity after treated with capsaicin were evaluated by electrophoretic mobility gel shift assay (EMSA). ESCC cells were treated with capsaicin, 5-FU or combinations thereof for 72h and then analyzed cell apoptosis by flow cytometry. RESULTS: Two ESCC cell lines had expression of IκBα and pIκBα in the cytoplasm, p50 and p65 in the nuclear. The results also indicated that capsaicin inhibited IκBα phosphorylation and Bcl-2 in two ESCC cell lines in a concentration-dependent manner. EMSA showed high DNA-binding activity in two ESCC cell lines and capasicin suppressed the NF-κB-DNA-binding in a concentration-dependent manner. Capsaicin alone could increase cell apoptosis (P<0.05). While combination with 5-FU, the results were more prominence (P<0.05). CONCLUSION: Capsaicin inhibits the phosphorylation of IκBα and downregulates the activation of NF-κB signaling pathway, leading to suppression of proliferation, induction of apoptosis and increasing the sensitivity of 5-FU in ESCC cell lines.
