calpain抑制剂（ALLN）对大鼠离体心脏缺血/再灌注损伤的保护作用

Objective :To evaluate calpain inhibitor (ALLN) mediate protective effect against ischemic-reperfusion(IR) injury of isolated rat hearts and its mechanism. Methods :Forty five rats were randomly separated into 5 groups: control group, IR group, control treated with ALLN ，IR treated with ALLN and IR treated with DMSO. Langendorff perfusion model was made in isolated hearts with IR injury, and investigate the effect of ALLN（10μM）on myocardial IR injury in rat hearts. coronary flux （CF）， the value of LDH，the protein content and activity of Calpain were investigated，The fluorescence intensity of intracellular Ca2+ and rate of apoptosis in rat cadiomyocytes were detected with FLUO-3/AM and Annexin-V FITC loading with flow cytometer，and then apoptotic indexes were calculated. Results: Compared with the IR group,those were significantly increased in CF and LDH（P < 0.01） ,but the protein content and activity of Calpain,、the fluorescence intensity of intracellular Ca2+ and the index of apoptosis were significantly low in IR treated with ALLN group（P < 0.01）. Conclusion: calpain inhibitor (ALLN) has a significant protective effect on IR injury and apoptosis of isolated hearts by preventing increase in activity of Calpain, which may be related with attenuating cadiomyocytes injury and lessening the intracellular calcium overload .
