目的:研究胶原蛋白肽对蛋氨酸限制高脂饮食小鼠体重、骨强度、骨组织氧化还原状态与微结构的影响。方法:36只C57BL/6雄性小鼠随机分为蛋氨酸限制组(MR)和胶原蛋白肽添加组(PMR)。在第22周时用综合实验动物监测系统(CLAMS)测定小鼠的能量代谢情况之后处死,取出股骨,扫描股骨微结构,测定股骨骨长度、骨干重、骨矿物质含量、骨密度、骨强度、骨组织氧化还原状态、成骨细胞和破骨细胞分化相关基因的表达。结果:与MR组小鼠相比,PMR组小鼠体重、能量支出、骨干重、骨矿物质含量、骨组织体积、骨密度、骨小梁数量、骨强度和单位骨强度都显著性降低(P<0.05),骨小梁分离度和破骨细胞分化相关基因表达显著性提高(P<0.05)。结论:饮食胶原蛋白肽添加会降低蛋氨酸限制高脂饮食小鼠的能量支出和骨矿物质含量,损害骨组织微结构,降低骨密度,从而降低骨强度,并且会破坏成骨细胞与破骨细胞之间的平衡。因此,在饮食蛋氨酸限制的条件下,添加胶原蛋白肽可能会给机体骨骼带来不利的影响。

Objective: To investigate the effects of collagen peptide on body weight, bone strength, bone tissue redox state and microstructure in mice fed with methionine restricted and high fat diet. Methods: 36 male C57BL/6 mice were randomly divided into methionine restricted diet(MR) and collagen peptide-methionine restricted diet(PMR). In the 22th week, mice energy metabolism was measured by comprehensive laboratory animal monitoring system(CLAMS) and then the mice sacrificed. Remove the femur, scanning femur microstructure, the length of femur, bone dry weight, bone mineral content, bone mineral density, bone strength, bone tissue redox state, the expression of osteoblast and osteoclast differentiation related genes were determined. Results: Compared with MR group, the body weight, energy expenditure, bone dry weight, bone mineral content, bone volume, bone mineral density, trabecular number, bone strength, bone strength/body weight significantly decreased(P<0.05). Trabecular separation and the osteoclast differentiation related genes significantly increased(P<0.05). Conclusion: The addition of dietary collagen peptide can reduce energy expenditure and bone mineral content, damage the microstructure of bone tissue, reduce bone mineral density, thereby reduce bone strength and destroy the balance between osteoblasts and osteoclasts in mice fed methionine restricted-high fat diet.Therefore, at the dietary methionine restriction, the addition of collagen peptides may have adverse effects on the mice bone.
