彗星电泳法测定双酚A对小鼠脑细胞的DNA损伤

目的:检测不同浓度双酚A长期暴露后对小鼠脑细胞的DNA损伤。方法:以茶油作为空白对照,分别以双酚A浓度为0、0.1μg/mL、10μg/mL和1000μg/mL的茶油灌胃雄性清洁级小鼠8周,然后利用彗星电泳法检测各组小鼠脑细胞的DNA损伤。结果:不同浓度双酚A暴露8周后,彗星电泳图像显示小鼠脑细胞DNA出现不同程度的损伤,随着暴露剂量的增加,带有彗尾的细胞比率从对照组的9.5%分别升高到暴露组的34.5%、36.0%和50.5%,细胞总体的尾部DNA含量、尾长和尾矩也都逐渐增加,而且各双酚A暴露组与空白对照组都具有显著性差异(p<0.01)。结论:长期双酚A暴露(包括低浓度环境暴露)会导致小鼠脑细胞的DNA损伤。

Objective: This study was aimed to investigate the DNA damages of brain cells in mice induced by long-term exposure to Bisphenol A. Methods: Four mice groups, including the control group and three exposure groups, chronically received different Bisphenol A treatments (0μg/mL, 0.1μg/mL, 10μg/mL and 1000μg/mL in tea oil) at Am 9:00-11:00 for 8 weeks. Then comet assay was employed to detect the DNA damages of the brain cells in these mice. Results: After the treatment with Bisphenol A, the comet percentages of brain cells increased from 9.5% in the control group to 34.5%, 36.0% and 50.5% in three exposure groups, respectively. And the percentages of tail DNA, tail length and tail moment in brain cells in the exposure groups were significantly higher than those in the control group (p< 0.01). Conclusion: The result showed that long-term exposure to Bisphenol A, including the low level exposure, could lead to DNA damages of the brain cells in mice and the damage levels increased with the increasing of Bisphenol A concentrations.
