OE1介导独脚金内酯调控拟南芥开花的机制研究
Study on the TOE1-mediated regulation mechanism of flowering by strigolactone in Arabidopsis
众所周知,开花调控是植物生长发育中重要的过程之一。一些植物激素,如赤霉素和生长素等在开花调控中发挥关键的作用。独脚金内酯(Strigolactones,SLs)是一种来源于β-胡萝卜素的萜类新型植物激素,具有调节植物生长和发育的重要作用。前期部分研究发现,SL在调控植物开花过程中可能发挥了一定的作用,但其确切作用及分子机制仍有待于进一步解析。本文以拟南芥为材料,通过对SL系列突变体进行表型鉴定,确认了内源SL抑制拟南芥开花的作用。然后,通过文库筛选获得了TOE1因子,其与SL受体D14及SL信号负调因子SMXL7均能够互作。通过对一系列多突及过表达突变体开花表型进行统计发现,TOE1是SL信号通路调控开花的关键因子,且其作用于SMXL6/7/8的下游。同时,SMXL7与TOE1互作抑制了其转录抑制活性,而D14则够增强其转录抑制活性,从而分别实现对下游FT基因的激活和抑制作用。本文明晰了SL调控植物开花时间的功能及相关分子机制,不仅丰富了SL信号转导调控机制,而且为揭示植物花期调控网络奠定了基础。
s is well known, flowering regulation is one of the important processes in plant growth and development. Some plant hormones, such as gibberellins and auxins, play crucial roles in flowering regulation. Strigolactones (SLs) are a novel class of terpenoid plant hormones derived from β-carotene, which play an important role in regulating plant growth and development. Previous studies have suggested that SLs may play a role in regulating plant flowering, but their exact function and molecular mechanism remain to be further elucidated. In this study, using Arabidopsis as the material, the phenotype identification of SL series mutants confirmed the inhibitory effect of endogenous SL on Arabidopsis flowering. Subsequently, through library screening, the TOE1 factor was identified, which interacts with the SL receptor D14 and the SL signaling repressor SMXL7. Statistical analysis of the flowering phenotypes of a series of multiple mutants and overexpression mutants revealed that TOE1 is a key factor in the SL signaling pathway regulating flowering and acts downstream of SMXL6/7/8. Furthermore, the interaction between SMXL7 and TOE1 inhibits its transcriptional repression activity, while D14 enhances its transcriptional repression activity, thereby respectively activating and inhibiting downstream FT genes. This study elucidates the function and related molecular mechanism of SL in regulating plant flowering time, not only enriching the understanding of SL signal transduction regulatory mechanisms but also laying the foundation for unraveling the plant flowering time regulation network.
