Rapid detection method for nitrofuran antibiotic residuesby surface-enhanced Raman Spectroscopy
硝基呋喃类抗生素是一类曾被广泛使用的兽药,由于其对人体的毒副作用已被各国禁止使用。本研究建立了一种表面增强拉曼光谱快速检测硝基呋喃类抗生素呋喃妥因、呋喃它酮及其混合物的方法。首先用Gaussian 03量子化学软件包,以B3LYP/6-31G(d)为方法,计算并分析了呋喃妥因及呋喃它酮分子的拉曼振动模式。其次用柠檬酸钠还原法制备了粒径约50nm左右的金溶胶,作为增强基底使用。由于呋喃妥因与呋喃它酮结构类似,故其用于定性的拉曼特征峰也相近,包括1602 cm-1,1556 cm-1,1484 cm-1,1336 cm-1,1008 cm-1和968 cm-1 ,而特征峰1420 cm-1,1388 cm-1和1456 cm-1,1242 cm-1,1162 cm-1则分别属于呋喃妥因与呋喃它酮,其中峰1420 cm-1 和1456 cm-1可用于鉴别二者的混合物。而峰1008 cm-1和1162 cm-1可分别用于呋喃妥因和呋喃它酮的定量检测。在最优检测条件下,二者的检测限为5 ppm。研究结果证明以金溶胶为增强基底的表面增强拉曼光谱法可以快速准确地鉴定呋喃妥因和呋喃它酮,为日后检测常规食品样品中的残留物提供了基础。
Nitrofurans are a group of widely used veterinary antibiotics which have been forbidden in many countries. Surface-enhanced Raman spectroscopy (SERS) coupled with gold nanoparticles was used in this study for rapid detection and characterization of nitrofuran antibiotics (including furadantin, furaltadone and their mixture). Density functional theory calculations was conducted with Gaussian 03 at the B3LYP level using the 6-31G(d) basis set to analysis the assignment of vibrations. The SERS measurement of nitrofuran antibiotics has been investigated by active SERS substrate of gold nanoparticles with average size around 50 nm. The SERS qualitative analyses has been studied for furadantin and furaltadone by their characteristic SERS pesks. The peaks at 1420 cm-1 and 1456 cm-1 could be used to discriminate their mixture. The SERS peaks at 1008 cm-1 and 1162 cm-1 were used as the index of quantitative analysis for furadantin and furaltadone, respectively. Under the optimal conditions, the limit of detection could reach the level of 5 ppm. The results showed that the SERS method with gold nanoparticles was capable of identifying and characterizing furaltadone and furadantin, which was valuable to be further applied on the detection of antibiotic residues in food sample.
