不结球白菜亮白叶再生体系的优化
Optimization of Regeneration System in vitro of non-heading Chinese Cabbage
为转基因工作建立良好的再生体系,以不结球白菜品种亮白叶为实验材料,研究不同外植体类型和不同激素配比等因素对其不定芽分化频率的影响。结果表明:对不结球白菜品种中再生比较困难的品种亮白叶而言,以下胚轴、子叶及子叶柄为外植体的诱导频率存在明显差异。以生长5~6天无菌苗的子叶柄为外植体,在培养基为MS+6-BA(5.0 mg/L)+NAA(0.1 mg/L)+AgNO3(4.0 mg/L)的培养条件下,亮白叶分化不定芽的再生频率最高,可达59.5%;这为将来该不结球白菜转化体系的建立奠定了基础。
o set up better regeneration system for transgenic work, the effects of explant type and hormone on the regeneration of the non-heading Chinese cabbage (Brassica campestris ssp. Chinensis Makino.) called Liangbaiye were discussed. The results showed that there was marked difference in the regeneration frequency between petiole with cotyledon, cotyledon and hypocotyl. The regeneration frequency of petiole with cotyledon was the highest among three explants\' types in the medium of MS + 6-BA (5.0 mg/L) + NAA (0.1 mg/L) + AgNO3 (4.0 mg/L), and the differentiation frequency could reach as high as 59.5%. The results layed the foundation for establishment of genetic transformation system of Liangbaiye.
