PH-2基因的交互作用与单相抑郁症关系

目的 探讨色氨酸羟化酶2(TPH-2)基因 rs4570625 和 rs11178997 的交互作用与单相抑郁症的关系。 方法 提取中国北方地区300例单相抑郁症患者(病例组)和300名健康对照者(对照组)基因组DNA,采用PCR技术分别扩增包含TPH-2基因 rs4570625 和 rs11178997 位点的基因组DNA片段,将PCR产物直接测序检测病例组及对照组的基因型,应用非条件logistic 回归模型分析 rs4570625 和 rs11178997 基因的交互作用。结果 病例组与对照组SNP(单核苷酸多态性)基因型分布符合Hardy-Weinberg 平衡定律(P>0.05);单相抑郁症组与健康对照组TPH-2基因rs4570625 位点基因型及等位基因频率比较,差异无统计学意义;rs11178997 基因型及等位基因频率2组比较差异均有统计学意义(均P<0.01),病例组携带的基因型G/G、A/G、A/A频率分别为6.0%、18.3%、75.7%,对照组分别为57.3%、2%、40.7%;交互作用分析显示,rs4570625 TG 基因型与 rs11178997 AG 基因型对单相抑郁症患病具有正交互作用(ORAB = 5.539, ORA = 0.747, ORB =0.024, ORI > 1)。 结论 rs4570625与 rs11178997 变异之间的交互作用在单相抑郁症发病机制中具有重要意义。
Objective To explore the significance of the interaction between tryptophan hydroxylase-2 (TPH-2) gene rs4570625 and rs11178997 in uniplor deression patients. Method We collected the specimens of peripheral blood from 300 uniplor depression patients and 300 healthy conrols in a mental hospital in Harbin city. PCR was used to detect the polymorphisms of rs4570625 and rs11178997, the logistic regression model was used to analyze the interaction of rs4570625 and rs11178997. Result the genotypic frequences of the SNP did not deviant from Hardy-Weinberg equilibrium in both case and control groups (P>0.05).Compared with the control group, no significant difference in genotypes and alleles of TPH2 gene rs4570625 was found in patient group(P>0.05). There was significant difference in G/G、A/G、A/A frequencies of TPH2 gene rs11178997 between the patient group (6.0%、18.3%、75.7%) and control group (57.3%、2%、40.7%). There was positive interaction between rs4570625 and rs11178997 in uniplor depression patients (ORAB = 5.539, ORA = 0.747, ORB =0.024, ORI > 1). Conclusion The interaction between rs4570625 and rs11178997 plays important role in uniplor depression patients.
