basta和卡那霉素是植物遗传转化研究中两种常用的抗性筛选剂,确定两者适当的选择压对于苎麻遗传转化研究有着重要意义。本文以湖北省主推品种"华苎4号"和"华苎5号"为材料,系统研究了basta和卡那霉素对其试管苗不同外植体再生的影响。结果表明:basta和卡那霉素均能显著降低"华苎4号"和"华苎5号"的再生频率,不同基因型的不同外植体的敏感性有所差别。对于"华苎4号"而言,最适宜的basta选择压为叶片0.5 mg/L、茎段0.6 mg/L、叶柄0.7 mg/L以上,最适的Km选择压为叶片25 mg/L;对于"华苎5号"而言,最适宜的basta选择压为叶片0.6 mg/L、茎段0.7 mg/L、叶柄0.7 mg/L,最适的Km选择压为叶片35 mg/L。

Basta and kanamycin (Km) were usually used as selection in plant transformation and it was of importance to identify their selection pressure before transgene. Huazhu No. 4 and Huazhu No. 5, mainly popularized varieties in Hubei province, were used to evaluate the effects of basta and Km on regeneration of different explants derived from test-tube plantlets in this study. The results showed that basta and Km significantly influenced plant regeneration ratio of Huazhu No. 4 and Huazhu No. 5, while different explants showed different reaction. To Huazhu No. 4, the optimum selection pressure of basta was 0.5 mg/L for leaves, 0.6 mg/L for stems and higher than 0.7 mg/L for petioles respectively and the optimum selection pressure of Km for leaves was 25 mg/L. But to Huazhu No. 5, the optimum selection pressure of basta was 0.6 mg/L for leaves, 0.7 mg/L for stems and petioles respectively and the optimum selection pressure of Km for leaves was 35 mg/L.
