低温光抑制后PSII活性的快速恢复不利于PSI活性的恢复
Faster recovery of PSII is detrimental to the recovery of PSI in cucumber (Cucumis sativus L.) leaves after chilling photoinhibition
为了探讨冷敏感植物黄瓜在低温弱光胁迫后常温恢复过程中光系统I(PSI)与光系统II(PSII)的恢复规律和相互作用,以及光照在此过程中的作用,以冷敏感黄瓜品种“津春4号”为试材,通过同时测定黄瓜叶片叶绿素荧光快速诱导动力学曲线和对820nm光的吸收曲线,结合叶绿素荧光淬灭分析,分析了低温(4℃)弱光(200μmol m-2 s-1)胁迫以及之后的常温(25℃)不同光强(0,15,200μmol m-2 s-1)以及有或没有DCMU存在情况下的恢复过程中,PSI和PSII活性的变化。结果表明,逆境消除后PSII最大光化学效率(Fv/Fm)立刻恢复且对光强不敏感。PSI活性(△I/Io)在常温15μmol m-2 s-1光下立刻恢复但恢复慢于Fv/Fm。在常温200μmol m-2 s-1光下,PSI活性恢复受到明显抑制,甚至继续下降,而DCMU预处理能有效的缓解了200μmol m-2 s-1光对PSI活性恢复的抑制作用。完全黑暗条件下两光系统均不能恢复。这表明恢复过程中减少PSII向PSI的线性电子传递能够避免PSI光抑制继续加重,加快PSI活性的恢复。PSII活性比PSI活性恢复快,这可能是导致PSII向PSI的线性电子传递过高的主要原因。在确定光强下,相对于PSI活性,PSII活性恢复越快,对PSI甚至整个光合机构活性的恢复越不利。
In order to investigated the effects of different light intensities (0, 15, 200 μmol m-2 s-1) with or without DCMU on recovery process of cucumber (Cucumis sativus L.) leaves after treatment with low temperature (6℃) combined with moderate light intensity (200 μmol m-2 s-1) for 6 h, chlorophyll a fluorescence transient and light absorbance at 820 nm as well as analyzing chlorophyll quenching were analyzed,. Both the Fv/Fm and the ΔI/Io decreased after chilling treatment under 200 μmol m-2 s-1 light. After the leaves were transferred to 25℃, Fv/Fm recovered immediately under both 200 and 15 μmol m-2 s-1 light. ΔI/Io recovered immediately under 15 μmol m-2 s-1 light, but the recovering rate of ΔI/Io was slower than that of Fv/Fm. The recovery of ΔI/Io was severely restrained by 200 μmol m-2 s-1 light, while a pretreatment with DCMU effectively alleviated this restraining. The results indicate that recrease of electron transport from PSII to PSI protected PSI from further photoinhibition, quicken up the recovery of PSI. Under a given light, faster recovery of PSII compare to PSI was badly for the recovery of PSI or even to the whole photosystem
