LC-3基因在宫颈癌中的表达及临床意义
Expression of CLC-3 mRNA in cervical cancer
目的 探讨容量调控性氯离子通道(CLC-3)在子宫颈癌组织中的表达及临床意义。方法 采用逆转录-聚合酶链反应(RT-PCR)方法检测60例宫颈癌组织,10例经放化疗前后的宫颈癌组织,10例正常宫颈组织中的CLC-3mRNA的表达情况。结果 CLC-3mRNA在正常宫颈组织中的表达的灰度值的比值为0.6912与宫颈癌组织中的2.3937相比,有显著性差异(P<0.01)。CLC-3mRNA在高分化癌中表达为1.3187、中分化为1.5718,与低分化的4.5899 相比,有显著性差异(P<0.05)。CLC-3mRNA在宫颈癌Ⅰ-Ⅱ期中表达为1.0842,在宫颈癌Ⅲ-Ⅳ期表达为3.5688,两者有显著性差异(P<0.01)。CLC-3基因在放化疗前的宫颈癌组织中表达为1.5198与放化疗后的0.2237相比,有显著性差异 (P<0.01)。结论 CLC-3基因参与宫颈癌细胞恶性增殖过程。CLC-3与宫颈癌的恶性进展有关、与宫颈癌的临床分期及病理分级呈正相关。宫颈癌放化疗可降低CLC-3基因的表达。
Objectives To study the expression of CLC-3 mRNA in cervical cancer. Methods Reverse transcription polymerase chain reaction(RT-PCR) was used to detect the expression of CLC-3 mRNA in 60 patients with cervical cancer,10 patients with cervical cancer before and after radiotherapy and chemotherapy, and 10 patients with normal cervical tissue. Results the ratio of gray level of CLC-3 mRNA was 0.6912 in normal cervical tissue, 2.3937 in cervical cancer, there were significant differences between them (P<0.01). The ratio of gray level of CLC-3mRNA was 4.5899 in the poor differentiation cancer, higher than that of the middle and high differentiation cancer 1.5718 and 1.3187 (P<0.05). the ratio of gray level of CLC-3mRNA was 1.0842 in stages Ⅰ~Ⅱ, 3.5688 in stages Ⅲ~Ⅳ, there were significant differences between them (P<0.01). CLC-3mRNA was 1.5198 before radiotherapy and chemotherapy, but 0.2237 after radiotherapy and chemotherapy, they have significant differences(P<0.01). Conclusion CLC-3mRNA participates in the proliferation of cervical cancer cell, and plays an important role in tumorgenesis of cervical cancer. CLC-3mRNA have a positive correlation with clinical stages and pathological grades of cervical cancer. The expression of CLC-3mRNA in cervical cancer was decreased after radiotherapy and chemotherapy.
