葛根素定量分析及对卵巢切除小鼠骨密度作用

目的:定量分析葛根中主要异黄酮成分,并观察其最主要异黄酮葛根素对卵巢切除小鼠骨量减少的保护作用。方法:HPLC对葛根中葛根素、大豆苷元、染料木素三种主要异黄酮进行定量分析。另外,实验设假手术组,模型组,葛根素低、中和高剂量组以及雌二醇组。除假手术组施行假手术外,其余各组小鼠都施行卵巢切除。术后葛根素低、中和高剂量组小鼠每天分别经口摄取2、4、8mg葛根素,雌二醇组被皮下给予17β-雌二醇0.03μg/day,4周后对小鼠股骨骨密度进行检测。结果:HPLC分析显示,葛根中异黄酮主要以葛根素为主,还含有存在于大豆的大豆苷元和染料木素,他们的含量分别为43.5mg/g, 8.03mg/g, 1.01mg/g。动物实验结果表明,卵巢切除4周后,小鼠股骨近端、中段、远端以及股骨全体骨密度均现下降,其中以股骨远端下降最为显著。葛根素各剂量组对股骨各部位和股骨全体骨密度下降均有一定抑制作用,其中以低剂量葛根素对股骨远端骨密度作用最为显著。雌二醇完全抑制了股骨各部位以及股骨全体骨密度的下降。结论:葛根中异黄酮以葛根素为主,还含有存在于大豆的二种主要异黄酮大豆苷元和染料木素。葛根素具有部分抑制卵巢切除所致小鼠骨量减少的作用,是葛根防治骨质疏松症重要的物质基础。

Objective: Radix Puerariae (RP), the root of Puerariae labata (Willd) Ohwi, a wild creeper leguminous plant, is an important crude edible herb and one of the earliest traditional chinese medicine for various medicinal purposes. In previous paper, we have reported that the overall crude herb of RP inhibited bone loss by low dose and increased bone mass by high dose in OVX mice. RP contains high level of isoflavones. In this paper, we analysed the main isoflavones in RP by HPLC and examined the role of major isoflavone Puerarin on bone protective effects in ovariectomized mice. Methods: To quantfy the total isoflavones, 10mg RP powder was mixed with 25μl β-glucuronidase solution in 200μl sodium acetate buffer and incubated for 37℃ for 3h,then 800μl menthanol was added and mixed well. After the sample was centrifuged, the supernate was passed through the 0.22μm micorfilter, and 10μl of the extract was applayed to HPLC. On the other hand, to examined the role of puerarin on bone metabolism, female mice were ovariectomized (OVX), and administration with low, middle and high dose of puerarin or 17-beta-estradiol, 4 weeks after the femoral bone mineral density (BMD) were measured. Results:HPLC chromatogram showed that puerarin is the major peak and it amounted to 43.5mg/g of the RP in dry weight. Daidzein and Genistein were other two peaks, and were 8.03mg/g and 1.01mg/g separately. The whole femoral bone mineral density (BMD) was significantly reduced by OVX, and the decrease in BMD was inhibited by any of thd dose of RP, and low dose of RP is significantly. Treatment with 17-beta-estradiol not only completely inhibited the decrease in BMD,but also increased the BMD in OVX mice. Conclusions: The isoflavones containing in RP are mainly puerarin, and also contanin two of the major isoflavones Daidzein and Genistein existed in soybean. Puerarin has shown inhibited bone loss in ovariectomized mice, and should be one of the important components responsble for regulating bone matabolism by RP.
