FLII蛋白抑制肺腺癌细胞H1299的迁移和侵袭
FLII protein inhibits migration and invasion of H1299 lung adenocarcinoma cell
肺腺癌细胞H1299具有较高的转移能力,有文献报道FLII参与细胞骨架重排以及细胞的迁移和侵袭,但其对肺腺癌细胞H1299转移能力的影响还未见报道。本研究中,我们首先在正常细胞、肺腺癌细胞、乳腺癌细胞株中检测FLII蛋白的表达水平,发现在肺腺癌H1299细胞株中FLII的表达量最低。通过构建的真核表达载体pCMV-N-Flag-FLII转染H1299细胞,以及利用G418,我们筛选获得稳定表达Flag-FLII的H1299细胞株。通过transwell实验证明FLII抑制H1299细胞的迁移和侵袭,这些结果为深入研究FLII的生物学功能奠定了基础。
H1299 is a highly invasive lung adenocarcinoma cell. FLII protein has been shown by literatures to be involved in cytoskeletal rearrangement and cell migration and invasion. However, its role in the migration and invasion of H1299 cells has not yet been reported. In this study, we firstly detected the expression level of FLII protein in normal cells, lung cancer cells, and breast cancer cells. Our results indicated that FLII expression in H1299 and A549 lung cancer cells was significantly lower than that in the control HBE cells. We have constructed the eukaryotic expression vector pCMV-N-Flag-FLII, and transfected it in H1299 cells. Then, By screening using G418, we established the H1299 cell line stably expressing Flag-FLII. Transwell experiments using these cells demonstrated that FLII could inhibit the migration and invasion of H1299 cells, which provides a useful tool for further study on the biological function of FLII protein.
