SPOI标记猪脂肪干细胞向成骨细胞分化及体外MRI成像
ifferentiation of SPIO labeling ADSCs from pigs to osteoblast and in vitro 3.0T MR imaging
目的:研究SPIO磁标记对猪脂肪干细胞向成骨细胞分化的影响以及磁标记脂肪干细胞的体外3.0T MR 成像特性。方法:实验小型猪皮下脂肪分离培养脂肪干细胞,超顺磁性氧化铁-多聚左旋赖氨酸复合物标记脂肪干细胞;对磁标记细胞及未标记细胞成骨诱导。应用3.0T MR 对不同浓度磁标记细胞进行T1WI、T2WI 及T2*WI序列体外成像。结果:普鲁士蓝染色显示磁标记细胞胞质内含有蓝染铁颗粒,细胞标记率近100%;磁标记脂肪干细胞向成骨细胞方向诱导分化成功,标记细胞与未标记细胞均可分化为成骨细胞并形成钙结节。MRI成像显示随细胞浓度升高,3 种序列信号变化率均增加,T2*WI信号变化率最大。3.0T MR 成像能检测到至少1×103 /mL 标记细胞。结论:脂肪干细胞是组织工程骨和骨移植的理想种子细胞。3.0T MR可敏感的检测磁标记干细胞。
OBJECTIVE: To study the effects of superparamagnetic iron oxide nanoparticles (SPIO) labeling on the differentiation potential of adipose tissue-derived stem cells (ADSCs) to osteoblasts, and the characteristics of magnetic labled ADSCs in vitro 3.0T magnetic resonance imaging(MRI). METHODS: ADSCs were isolated from the subcutaneous adipose tissue of experimental pigs and expanded in culture medium. ADSCs were incubated with the SPIO-PLL complex. The differentiation capacity of ADSCs to osteogenic lineage between labeled and unlabeled cells was compared. Different concentrations of labeled ADSCs and the control were scanned at a 3.0T MR system with T1WI , T2WI and T2*WI. RESULTS: Numerous intracytoplasmic iron particles stained with Prussian blue were detected in labeled cells with labeling efficiency of nearly 100%. Labled ADSCs were capable of differentiating to osteoblasts. Both labeled and unlabeled cells were able to differentiate into osteoblasts and calcification formation was found in both osteogenic lineage. The change in MR signal intensity increased at T1WI, T2WI and T2*WI along with elevation of the labeled cells concentration. The marked change in signal intensity were observed at T2*WI . The lowest concentration of SPIO labeled ADSCs detected by 3.0T MRI was 1×106/L cell. CONCLUSION: ADSCs are promising cells in bony tissue engineering and bony grafting.SPIO labled ADSCs could be traced by 3.0T MR.
