古细菌细胞壁表面的S-层蛋白具有独特的自组装特性,能够在界面上重新自组装形成纳米级二维网格结构,近年来已经成为纳米领域的研究热点。而深入研究S-层蛋白的分子结构将有助于揭示其自组装机理,扩大S-层蛋白自组装结构的应用领域。制备S-层蛋白有序结晶是研究其分子结构研究的重要基础,本文从芽孢杆菌中提取S-层蛋白并观察了其自组装。此外,成功在硅烷化的玻璃表面构建了生物素-亲和素(biotin-avidin)分子链接,促进S-层蛋白在固基表面的自组装与结晶,成功得到了S-层蛋白的规则晶体,为S-层蛋白的三维结构分析提供了研究基础,推动了S-层蛋白自组装及改性机理研究的进展。

S-layer protein has the unique ability to re-assemble into two-dimensional monolayer grid structure and in recent years it has attracted great attention in the field of biomimetic and nanotechnology. The ordered crystalline S-layer protein is an important foundation to study the structure of the protein molecular, we successfully obtained S-layer protein crystal using the silanized glass surface modified by biotin-avdin. These nice protein crystals are expected to contribute to the three-dimensional structure analysis of the protein and promote the progress of the research on self-assembly and modification of S- layer protein.
