纤维强度不同的品种在棉纤维发育过程中基因表达差异的分析

本研究以陆地棉(五个高强以及两个对照品种)和海岛棉的纤维为材料,利用cDNA-AFLP技术分析了纤维快速伸长期、纤维伸长与次生壁增厚重叠期(简称重叠期)、次生壁快速增厚期其基因表达的差异情况。利用256对选择性扩增扩引物组合共获得了各种类型差异条带271条。对其中的16条进行回收测序,通过Blastx比对发现其中3条与纤维素合酶、Fb8-like mRNA、LTP家族基因等棉纤维发育基因的同源性分别为84%、98%以及99%,其他序列与已知棉花基因无明显同源性,推测为未知的棉纤维发育的基因片段。
In this experiment,we employed cDNA-AFLP for studying genes expression differences among phases for rapid elongation, overlapping and secondary wall rapid thickening of fiber by comparing five cotton(Gossypium hirsutum L.) cultivares of high-strength fibers (2001,7235,Beiersinuo, Acala1517-77, PD5264) with one cultivar of nomal fiber(shiyuan321, meifuchangrong ) and one cultivar of high- strength and thin fibers (hai7124)(Gossypium barbadense L.). we accomplished the analysis of gene expression difference covering 256 pairs of selective primers ,and got 271 ESTs. Sixteen fragments were cloned and sequenced,by BlastX in NCBI and fonud three of them is 84% homologous with cellulose synthase mRNA in cotton fiber; 98% homologous with fiber protein Fb8-like mRNA; 99% homologous with lipid transfer protein(LTP)mRNA; the rest are unknown proteins or lack of homologous, we speculate that they are new gene fragments in development of cotton fiber.
